摘要
目的测定不同产地厚朴中厚朴酚与和厚朴酚的含量,观察不同产地厚朴入药后对栀子厚朴汤特征成分溶出的影响,探究中药饮片化学成分差异对入药后复方汤剂化学轮廓的影响。方法建立10批厚朴饮片及其入药后栀子厚朴汤的高效液相色谱(HPLC)指纹图谱,通过对照品外标法测定其中厚朴酚与和厚朴酚的含量,以及栀子厚朴汤中主要成分栀子苷、柚皮苷、橙皮苷、芸香柚皮苷、新橙皮苷、橙皮素、柚皮素、厚朴酚、和厚朴酚的质量浓度。结果主成分分析(PCA)结果表明,10批厚朴质量具有明显差异,正交偏最小二乘判别分析(OPLS-DA)贡献率结果表明,和厚朴酚含量是厚朴质量差异最主要的影响因素;栀子厚朴汤中特征成分的溶出受厚朴不同产地的影响;灰色关联度分析结果表明,和厚朴酚与各成分溶出度关联度最大,且随着厚朴中和厚朴酚含量的增加,各成分的溶出有不同程度的降低。结论为有效控制厚朴饮片质量、栀子厚朴汤的抗抑郁药效及可能的毒性,应密切关注厚朴中和厚朴酚的含量。
Objective To determine the content of magnolol and honokiol in Magnolia officinalis Rehd. Et Wils. from different producing areas,observe the effect of Magnolia officinalis Rehd. Et Wils. used as medicine from different producing areas on the dissolution of component of Zhizi Houpo Tang(栀子厚朴汤),and explore the effect of chemical composition difference of Chinese herbal pieces on the chemical profile of compound decoction used as medicine.Methods High performance liquid chromatography(HPLC) fingerprint of 10 batches of herbal pieces of Magnolia officinalis Rehd. Et Wils. and Zhizi Houpo Tang thereof used as medicine were established,the content of magnolol and honokiol in Zhizi Houpo Tang as well as the mass concentrations of geniposide,naringin,hesperidin,narirutin,neohesperidin,hesperetin,naringenin,magnolol and honokiol therein were determined by reference substance along with external standard method. Results The results of principal component analysis(PCA) showed that there were significant differences in the quality of 10 batches of Magnolia officinalis Rehd. Et Wils. The contribution rate of orthogonal partial least squares discriminate analysis(OPLS-DA) showed that the content of honokiol was the most important factor affecting the quality difference of Magnolia officinalis Rehd. Et Wils;the dissolution of characteristic components in Zhizi Houpo Tang was affected by Magnolia officinalis Rehd. Et Wils. from different producing areas;the grey correlation analysis showed that the correlation between honokiol and dissolution of each component was the highest,with the increase of the content of honokiol in Magnolia officinalis Rehd. Et Wils.,the dissolution of each component decreased to some extent. Conclusion In order to effectively control the quality of Magnolia officinalis Rehd. Et Wils. herbal pieces,the antidepressant effect and possible toxicity of Zhizi Houpo Tang,we should pay close attention to the content of honokiol in Magnolia officinalis Rehd. Et Wils.
