菌核侧耳菌核水提物对对乙酰氨基酚诱导小鼠急性肝损伤的保护作用 被引量:1

摘要 研究了菌核侧耳(Pleurotus tuber-regium)菌核低、中、高剂量水提物(PTRs)(剂量分别为100、200、400 mg·kg^-1)对对乙酰氨基酚(acetaminophen,APAP)诱导C57BL/6小鼠急性肝损伤的保护作用。结果表明:与模型组比较,PTRs各剂量组小鼠血清中谷丙转氨酶(alanine aminotransferase,ALT)、乳酸脱氢酶(lactate dehydrogenase,LDH)含量极显著下降,中、高剂量组谷草转氨酶(aspartate aminotransferase,AST)含量极显著下降,高剂量组碱性磷酸酶(alkaline phosphatase,AKP)含量极显著下降。PTRs中、高剂量组小鼠肝脏中丙二醛(malondialdehyde,MDA)含量极显著降低,超氧化物歧化酶(superoxide dismutase,SOD)含量极显著升高;高剂量组谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)含量极显著升高。PTRs可以改善小鼠肝细胞脂肪性病变、坏死及炎性浸润,并明显减轻肝细胞损伤。提示PTRs能够显著提高小鼠的抗氧化能力,对APAP所致的急性肝损伤具有明显的保护作用。 The protective effect of water extract from Pleurotus tuber-regium sclerotia(PTRs)on acute liver injury induced by acetaminophen(APAP)in C57 BL/6 mice was investigated.Mice were randomly divided equally into six groups as follows:normal control group,model group,positive control group,low dose PTRs group(100 mg·kg^-1),medium dose PTRs group(200 mg·kg^-1),and high dose PTRs group(400 mg·kg^-1).Mice in PTRs groups were administered different doses of PTRs intraperitoneally,mice in the positive group were administered N-acetyl-L-cysteine(NAC)solution intraperitoneally,and mice in the normal control group and model group were administered PBS solution intraperitoneally.Compared with the model group,serum alanine transaminase(ALT)and lactate dehydrogenase(LDH)levels in all three PTRs groups,serum aspartate aminotransferase(AST)level in both medium and high dose groups,and serum alkaline phosphatase(AKP)level in the high dose group were reduced significantly.In addition,mouse liver malondialdehyde(MDA)contents in both medium and high dose groups were significantly decreased compared with the model group.In contrast,superoxide dismutase(SOD)activity in both medium and high dose groups and glutathione peroxidase(GSH-Px)activity in the high dose group were significantly increased compared with the model group.PTRs ameliorated the hepatocyte steatosis,necrosis,and inflammatory infiltration,suggesting that PTRs alleviated the acute liver injury induced by APAP through improving the antioxidant ability in mice.
