摘要
目的探讨异丙酚改善氯胺酮诱发的老龄大鼠认知功能障碍的机制。方法选择64只18月龄的健康雄性SD大鼠,依照随机数字表法分为对照组(C组)、异丙酚组(Y组)、氯胺酮组(L组)和异丙酚+氯胺酮组(Y+L组),各16只大鼠。Y组大鼠予以10 mg/kg异丙酚麻醉诱导,静脉输注异丙酚30 mg/(kg·h)维持;L组大鼠予以10 mg/kg氯胺酮麻醉诱导,静脉输注氯胺酮40 mg/(kg·h)维持;Y+L组大鼠予以10 mg/kg异丙酚+10 mg/kg氯胺酮麻醉诱导,静脉输注异丙酚30 mg/(kg·h)+氯胺酮40 mg/(kg·h)维持;C组大鼠予以等量0.9%氯化钠溶液处理。所有大鼠每天麻醉维持2 h,连续7 d。每天麻醉苏醒后采用Morris水迷宫测试和旷场实验分析各组大鼠认知功能。之后立即处死大鼠,每组中8只进行海马组织白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平检测,余下8只进行海马组织CA1区神经元凋亡和caspase-3表达水平检测。结果L组和Y+L组大鼠第1~5天时的Morris水迷宫测试逃避潜伏期以及旷场实验中央格停留时间均明显高于C组和Y组,穿越原平台所在区域的次数和时间以及跨格次数和直立次数均明显少于C组和Y组,海马区IL-6、TNF-α水平以及CA1区神经元凋亡百分比和caspase-3表达阳性神经元百分比均明显高于C组和Y组,差异有统计学意义(P<0.05)。Y+L组大鼠第1~5天时的Morris水迷宫测试逃避潜伏期以及旷场实验中央格停留时间均明显低于L组,穿越原平台所在区域的次数和时间以及跨格次数和直立次数均明显多于L组,海马区IL-6、TNF-α水平以及CA1区神经元凋亡百分比和caspase-3表达阳性神经元百分比均明显低于L组,差异有统计学意义(P<0.05)。C组与Y组大鼠第1~5天时的Morris水迷宫测试逃避潜伏期、穿越原平台所在区域的次数和时间,旷场实验中央格停留时间、跨格次数和直立次数,海马区IL-6、TNF-α水平以及海马区CA1区神经元凋亡百分比和caspase-3表达阳�
Objective To explore the mechanism of propofol in improving the cognitive impairment of aged rats induced by ketamine.Methods Sixty four 18 month old healthy male SD rats were randomly divided into control group(group C),propofol group(group Y),ketamine group(group L)and propofol+ketamine group(group Y+L),16 rats in each group.Group Y rats were anesthetized with 10 mg/kg propofol and maintained by intravenous infusion of 30 mg/(kg·h)propofol;group L rats were anesthetized with 10 mg/kg ketamine and maintained by intravenous infusion of 40 mg/(kg·h)ketamine;group Y+L rats were anesthetized with 10 mg/kg propofol and 10 mg/kg ketamine and maintained by intravenous infusion of 30 mg/(kg·h)propofol and 40 mg/(kg·h)ketamine;group C rats were anesthetized with the same amount of physiological salt Water treatment.All rats were anesthetized for 2 hours a day for 7 days.Morris water maze test was used to analyze the cognitive function of rats in each group.After that,the rats were killed immediately.The levels of IL-6,TNF-α in hippocampus were detected in 8 rats in each group.The apoptosis and Caspase-3 expression of neurons in CA1 area were detected in the remaining 8 rats.Results The escape latency of Morris water maze test and central grid dwell time in open field experiment in L group and Y+L group were significantly higher than that in C group and Y group,the times and times of crossing the original platform area,cross grid times and vertical times were significantly shorter than those in C group and Y group,the levels of IL-6,TNF-α,the percentage of apoptosis and Caspase-3 expression positive neurons in CA1 area were significantly higher than those in C group and Y group,the differences were statistically significant(P<0.05).The 1 to 5 d escape latency of Morris water maze test and central grid dwell time in open field experiment in Y+L group were significantly lower than thsoe in L group,and the times and times of crossing the original platform area,cross grid times and vertical times were significantly long
