摘要
基于GC-MS研究蓝莓花青素对镉致小鼠心脏损伤的保护作用并探讨其作用机制.将40只雄性昆明小鼠随机分为空白组、模型组及蓝莓花青素低、中、高剂量组(0.3 mg/kg/d、3 mg/kg/d、30 mg/kg/d),每组8只.除空白组外,其余各组小鼠腹腔注射CdCl 2构建镉损伤小鼠模型,蓝莓花青素各保护组小鼠灌胃不同剂量蓝莓花青素进行保护,连续处理14 d.采用GC-MS技术分析各组小鼠心脏组织脂质代谢物,运用SIMCA-P11.0进行多元统计分析.通过主成分分析(PCA)得分图可以发现各组有一定程度区分,采用偏最小二乘法(PLS)进一步分型,各组区分明显,蓝莓花青素保护效果呈梯度相关.与空白组相比,模型组小鼠心脏组织亚油酸、二十碳五烯酸、二十二碳六烯酸含量下降,其中二十二碳六烯酸含量下降最多,含量仅为空白组的62%,棕榈酸、硬脂酸、花生四烯酸含量增加,其中硬脂酸含量增加最多,含量为空白组的3.19倍,与模型组相比,蓝莓花青素保护组小鼠各项脂肪酸含量趋向正常组.说明蓝莓花青素通过调节脂质代谢紊乱,进而改善镉毒性造成的小鼠心脏损伤.
The protective effect of blueberry anthocyanin on cadmium-induced heart injury in mice was investigated and its mechanism was discussed based on GC-MS method.Forty male Kunming mice were randomly divided into five groups,including blank group,model group and three blueberry anthocyanin-protective groups with different doses(0.3 mg/kg/d,3 mg/kg/d,30 mg/kg/d),8 mice in each group.Cadmium-exposed mouse model were constructed by intraperitoneally injected with CdCl 2 except in blank group.Meanwhile,different doses of blueberry anthocyanin(0.3 mg/kg/d,3 mg/kg/d,30 mg/kg/d)were given to assess the protective effect of blueberry anthocyanin for 14 days.The lipid metabolites profiles in mice heart tissue of each group were analyzed using GC-MS,and multivariate statistical analysis were performed using SIMCA-P11.0.As shown in Principal component analysis(PCA)score map,it can be found that each group has a certain degree of differentiation.Partial least squares(PLS)was used for further classification.Each group is clearly distinguished,and the protective effect of blueberry anthocyanin is in a dose-dependent manner.Compared with the blank group,the content of linoleic acid,eicosapentaenoic acid,and docosahexaenoic acid was decreased(P<0.05)in the model group,among them,the content of docosahexaenoic decreased the most,and the content was only 62%of the blank group;the content of palmitic acid,stearic acid,and arachidonic acid were increased in the model group,among them,the stearic acid content increased the most,and the content was 3.19 times of the blank group.However,protective with blueberry anthocyanin can ameliorated the change of these lipid levels.Our data showed that blueberry anthocyanins can regulate the disorder of lipid metabolism and then exert certain protective effect against the heart injuries induced by cadmium intoxication.
