摘要
目的:探讨芸香苷对创伤性脊髓损伤(SCI)小鼠的影响及作用机制。方法:将40只C57BL/6小鼠随机分为假手术组、模型组、芸香苷组和强的松组,每组10只。除假手术组外,其他4组均复制创伤性SCI模型。建模后2 h,芸香苷组和强的松组小鼠分别腹腔注射芸香苷(100 mg/kg)、强的松(50 mg/kg),连续7 d,假手术组和模型组注射等量生理盐水。术后第1、第3、第7天,采用Basso Beattie&Bresnahan(BBB)评分观察小鼠SCI后后肢运动功能变化,苏木精-伊红(HE)染色法观察脊髓组织病理学变化,尼氏染色和TUNEL染色分别检测脊髓神经细胞表达与凋亡情况,酶联免疫吸附测定(ELISA)法检测白细胞介素(IL)-1β、IL-6和肿瘤坏死因子-α(TNF-α)表达,免疫荧光染色法检测激活转录因子核因子E2相关因子2(Nrf2)表达,Western blotting法分析Nrf2、血红素加氧酶-1(HO-1)蛋白表达。结果:术后第3天和第7天,芸香苷组和强的松组BBB评分较模型组明显升高(P<0.05)。与假手术组比较,模型组小鼠脊髓组织结构出现明显损伤,IL-1β、TNF-α、IL-6含量升高,尼氏体数目明显减少,神经细胞凋亡数目增加,Nrf2绿色荧光强度增加,Nrf2、HO-1蛋白表达增加;而芸香苷组和强的松组小鼠脊髓组织病变明显改善,并抑制了IL-1β、TNF-α、IL-6含量的增加,而尼氏体数目明显增加,同时减少了神经细胞的凋亡,Nrf2绿色荧光强度更强,脊髓组织中Nrf2、HO-1蛋白表达进一步增加(P<0.05);芸香苷组与强的松组作用情况比较,差异无统计学意义(P>0.05)。结论:芸香苷对创伤性SCI小鼠损伤脊髓有保护作用,其机制可能抑制炎症反应、激活Nrf2/HO-1途径相关。
Objective:To explore the effect and mechanismof rutin on traumatic spinal cord injury(SCI)in mice.Methods:A total of 40 C57BL/6 mice were randomly divided into sham operation group,model group,rutin group(n=10)and prednisone group,with 10 mice in each group.Except for the sham operation group,the traumatic SCI model was replicated in the other 4 groups.Two hours after the establishment of the model,the mice in the rutin group and prednisone group were intraperitoneally injected with rutin(100 mg/kg)and prednisone(50 mg/kg)for 7 days,and the sham operation group and the model group were injected with the same amount of normal saline.On the 1st,3rd and 7th day after operation,Basso Beattie&Bresnahan(BBB)score was used to observe the changes of hindlimb movements after SCI.HE staining was used to observe the histopathological changes of spinal cord.Nissl staining and TUNEL staining were used to detect the expression and apoptosis of spinal cord nerve cells.The expression of IL-1β,IL-6 and tumor necrosis factor-α(TNF-α)was detected by ELISA.The expression of nuclear factor E2-related factor 2(Nrf2)was detected by immunofluorescence staining.The expression of Nrf2 and heme oxygenase-1(HO-1)protein was analyzed by Western blotting.Results:On the 3rd and 7th day after operation,the BBB scores of rutin group and prednisone group were significantly higher than those of model group(P<0.05).Compared with the sham operation group,the spinal cord tissue of the model group showed obvious injury,the contents of IL-1β,TNF-αand IL-6 increased,the number of Nissl bodies decreased,the number of nerve cell apoptosis increased,the green fluorescence intensity of Nrf2 increased,and the expression of Nrf2 and HO-1 protein increased.However,the pathological changes of spinal cord tissue in rutin group and prednisone group were significantly improved,and the contents of IL-1β,TNF-αand IL-6 were inhibited,while the number of Nissl bodies was significantly increased,and the apoptosis of nerve cells was decreased.The green fluorescen
