摘要
目的分析乳腺癌组织中E2F家族蛋白的表达特征,并探讨E2F家族蛋白与乳腺癌患者预后的关系。方法从癌症基因组图谱数据库查询公开的乳腺癌组织与癌旁组织表达量数据,样本共891例,将样本按照分子分型标准分为Luminal A型438例、Luminal B型122例、HER-2型134例和TNBC型118例,另有癌旁组织样本79例。应用SPSS 22.0软件对不同亚型乳腺癌组织中E2F家族蛋白表达量与癌旁组织的差异进行统计学分析,并分析E2F家族蛋白与乳腺癌生存预后的相关性。结果 E2F1、E2F2、E2F3、E2F5、E2F7和E2F8蛋白在乳腺癌组织中呈高表达,在癌旁组织中呈低表达,两组比较差异显著(t=22.318、10.710、16.755、11.179、15.967、22.069,P均<0.001)。除E2F2蛋白表达在Luminal A组和Luminal B组比较差异不显著外(P>0.05),其他蛋白在各亚型组间两两比较差异均显著(P<0.05)。E2F1、E2F2、E2F3、E2F5、E2F7和E2F8蛋白表达量与患者生存预后呈明显相关性(P均<0.01),各蛋白的表达量越高,则患者的预后越差,生存率越低。结论 E2F家族蛋白中的E2F1、E2F2、E2F3、E2F5、E2F7和E2F8蛋白在乳腺癌组织中呈高表达,其表达水平与乳腺癌患者的发生发展及生存预后密切相关,可以为乳腺癌分子诊断与靶向治疗提供参考。
Objective To analyze the expression characteristics of E2F family proteins in breast cancer tissues and the relationship between E2F family proteins and prognosis of breast cancer patients. Methods A total of 891 samples of breast cancer tissues and adjacent tissue expression data were downloaded from the Cancer Genome Atlas database. The samples were divided into Luminal A type 438 cases,Luminal B type 122 cases,and HER-2 type 134. There were 118 cases with TNBC type and 79 cases with adjacent cancer tissue samples. SPSS 22. 0 software was used to statistically analyze the differences between the expression of E2F family proteins in different subtypes of breast cancer tissues and adjacent tissues,and to analyze the correlation between E2F family proteins and breast cancer survival prognosis.Results E2F1,E2F2,E2F3,E2F5,E2F7,and E2F8 proteins were highly expressed in breast cancer tissues and lowly expressed in adjacent tissues,and the differences were statistically significant(t = 22. 318,10. 710,16. 755,11. 179,15. 967,22. 069,P<0. 001). Except that the expression of E2F2 protein was not statistically significant between the Luminal B and Luminal A group(P> 0. 05),the other differential proteins were statistically significant in each pair(P< 0. 05). E2F1,E2F2,E2F3,E2F5,E2F7,and E2F8 protein expression levels were significantly correlated with patient’survival(all P<0. 01). The higher the expression of each protein,the worse the patient’s prognosis and the lower the survival rate.Conclusion E2F1,E2F2,E2F3,E2F5,E2F7 and E2F8 proteins in E2F family proteins are highly expressed in breast cancer tissues,and their expression levels are closely related to the development,survival and prognosis of breast cancer patients,which can provide reference for molecular diagnosis and targeted treatment of breast cancer.
