姜黄素对高糖波动下视网膜色素上皮细胞的保护作用

摘要 目的研究姜黄素对波动高糖条件下视网膜色素上皮细胞(RPE)的保护作用。方法将正常培养的ARPE-19细胞株分为正常组(NG)、波动高渗组(OC)、高糖组(HG)、波动高糖组(IHG)、血糖波动+低浓度给药组(10μg/mL姜黄素)、血糖波动+中浓度给药组(20μg/mL姜黄素)、血糖波动+高浓度给药组(40μg/mL姜黄素)。3d后观察各组细胞形态、细胞增殖活力、细胞内活性氧(ROS)水平。结果NG、HG、IHG 3组细胞活力依次降低(P<0.05),OC组与NG组无显著性差异(P均>0.05),给药组随着姜黄素浓度增加细胞活力增加,较IGH组显著增加(P>0.05)。NG、HG、IHG 3组细胞内ROS水平依次升高(P<0.05),OC组与NG组无显著性差异(P>0.05),给药组ROS水平随药物浓度增加而降低(P>0.05)。结论姜黄素可有效抑制波动高糖条件下的ARPE-19细胞氧化应激及提高细胞活力。 Objective To study the protective effect of curcumin on injury of retinal pigment epithelial cells induced by fluctuating high glucose.Methods Normal cultured ARPE-19 cells were divided into normal glucose group(NG),osmotic control group(OC),high glucose group(HG),intermittent high glucose group(IHG),intermittent high glucose+low dose group(10μg/mL),intermittent high glucose+medium dose group(20μg/mL),intermittent high glucose+high dose group(40μg/mL).After 3 days,cell morphology,cell proliferation activity and intracellular reactive oxygen specie(ROS)levels were observed.Results The cell viability was decreased and intracellular ROS level was elevated in HG and IHG groups as compared to NG group(P<0.05),and there was no significant difference between OC and NG group(P>0.05).With the increase of curcumin concentration,the cell activity was increased and ROS level was reduced in drug groups,compared with IGH group(P<0.05).Conclusion The curcumin can effectively inhibit the oxidative stress and improve cell vitality of ARPE-19 cells under the condition of fluctuating high glucose.
