前列腺组织中低氧诱导型转录因子-1α和可溶性转铁蛋白受体与前列腺癌病理特征的关系 被引量:4

Correlations of hypoxia-inducible transcription factor-1α and soluble transferrin receptor in prostate tissue with pathological characteristics of prostate cancer

摘要 目的观察前列腺组织中低氧诱导型转录因子-1α(hypoxia-inducible transcription factor-1α, HIF-1α)和可溶性转铁蛋白受体(soluble transferrin receptor, sTfR)的表达情况,探讨其与前列腺癌病理特征的相关性。方法取80例前列腺癌患者(前列腺癌组)手术切除的癌组织及癌旁正常组织、80例前列腺增生症患者(前列腺增生组)手术切除的前列腺增生组织,采用免疫组织化学法检测前列腺癌组织、前列腺增生组织、癌旁正常组织中HIF-1α、sTfR表达水平和阳性表达率;比较前列腺癌组不同Gleason分级患者HIF-1α、sTfR表达水平;采用Spearman相关性分析前列腺组织中HIF-1α、sTfR表达水平与前列腺癌病理特征的相关性。结果前列腺癌组织、前列腺增生组织HIF-1α[(87.50±1.50)%、(14.25±1.10)%]、sTfR[(88.00±1.45)%、(12.35±1.15)%]水平高于癌旁正常组织[(5.35±1.05)%、(6.20±1.20)%](P<0.05),前列腺癌组织高于前列腺增生组织(P<0.05);前列腺癌组织、前列腺增生组织HIF-1α(95.00%、20.00%)、sTfR(97.50%、25.00%)阳性表达率高于癌旁正常组织(5.00%、8.75%)(P<0.05),前列腺癌组织高于前列腺增生组织(P<0.05)。前列腺癌组Gleason G2级、G3~4级、G5级患者前列腺组织中HIF-1α[(70.30±3.85)%、(86.85±4.40)%、(98.25±4.25)%]、sTfR[(72.35±4.10)%、(87.50±3.25)%、(98.00±2.50)%]水平依次升高(P<0.05);前列腺组织HIF-1α、sTfR表达水平与前列腺癌Gleason分级均呈正相关性(r=0.660,P=0.001;r=0.710,P=0.001)。结论前列腺癌组织HIF-1α、sTfR呈高表达,其表达水平升高与前列腺癌的发生、发展具有相关性。 Objective To investigate the expressions of hypoxia-inducible transcription factor-1α(HIF-1α) and soluble transferrin receptor(sTfR) in prostate tissues and their correlations with the pathological characteristics of prostate cancer. Methods The surgically resected cancer tissues and adjacent normal tissues from 80 patients with prostate cancer(prostate cancer group) and the resected prostatic hyperplasia tissues from 80 patients with prostatic hyperplasia(prostatic hyperplasia group) were detected the expressions and positive rates of HIF-1α and sTfR by immunohistochemical method. The expressions of HIF-1α and sTfR in patients with different Gleason grades in prostate cancer group were compared.Spearman correlation analysis was used to study the correlations of the levels of HIF-1α and sTfR with the pathological characteristics of prostate cancer. Results The levels of HIF-1α and sTfR were higher in prostate cancer tissues((87.50±1.50)%,(88.00±1.45)%) and prostatic hyperplasia tissues((14.25±1.10)%,(12.35±1.15)%) than those in adjacent normal tissues((5.35±1.05)%,(6.20±1.20)%)(P<0.05), and higher in prostate cancer tissues than those in prostatic hyperplasia tissues(P<0.05). The positive rates of HIF-1α and STfR were higher in prostate cancer tissues(95.00%, 97.5%) and prostatic hyperplasia tissues(20.00%, 25.00%) than those in adjacent normal tissues(5.00%, 8.75%)(P<0.05), and higher in prostate cancer tissues than those in prostatic hyperplasia tissues(P<0.05). The levels of HIF-1α((70.30±3.85)%,(86.85±4.40)%,(98.25±4.25)%) and sTfR((72.35±4.10)%,(87.50± 3.25)%,(98.00±2.50)%) increased gradually in turn in patients with Gleason G2,G3-4 and G5 in prostate cancer group(P<0.05).The HIF-1αand sTfR levels in prostate tissues were positively correlated with the Gleason grade of prostate cancer(r=0.660,P=0.001;r=0.710,P=0.001).Conclusion HIF-1αand sTfR are highly expressed in prostate cancer tissues,which is correlated with the occurrence and development of prostate cancer.
