摘要 目的:通过网络药理学探索马齿苋治疗溃疡性结肠炎的作用机制。方法:利用TCMSP数据库查找马齿苋活性成分,借助PubChem,PharmMapper数据库预测所选活性成分作用靶点,运用TTD、Omim数据库收集溃疡性结肠炎靶点。通过STRING数据库和Cytoscape软件构建马齿苋活性成分-溃疡性结肠炎共同靶点蛋白互作网络并进行拓扑分析,筛选出马齿苋治疗溃疡性结肠炎的潜在靶点。利用KOBAS数据库、DAVID数据库进行潜在靶点的GO、KEGG富集分析。结果:筛选出马齿苋活性成分9个,马齿苋活性成分与溃疡性结肠炎共同靶点7个;潜在靶点GO富集涉及生物过程117条,细胞组分27种,分子功能31种;潜在靶点KEGG富集包含mTOR信号通路、PI3K-Akt信号通路等。结论:马齿苋治疗溃疡性结肠炎具有多分子、多靶点的特点,可能主要通过调节mTOR信号通路实现其治疗作用。 Objective:To explore the mechanism of Machixian(Portulacae Herba)in the treatment of ulcerative colitis(UC)by network pharmacology.Methods:The active components of Machixian(Portulacae Herba)were searched in the TCMSP database.The targets of the selected active ingredients was predicted by searching PubChem and PharmMapper databases,and the targets of UC was predicted by searching TTD and OMIM databases.Using the String database and Cytoscape software,the common target protein interaction network of the Machixian(Portulacae Herba)active ingredient-UC was constructed and topological analysis was carried out to screen out the potential targets of Machixian(Portulacae Herba)in the treatment of UC.The GO and KEGG enrichment analysis of potential targets was performed using the KOBAS database and the DAVID database.Results:There were 9 active constituents of Machixian(Portulacae Herba),7 common targets of Machixian(Portulacae Herba)active ingredients-UC.There were 117 biological processes,27 cellular components and 31 molecular functions predicted through GO enrichment for potential targets.According to KEGG enrichment for the potential targets,mTOR and PI3K-Akt signaling pathways were predicted.Conclusion:The treatment of UC with Machixian(Portulacae Herba)was multi-molecules and multi-targets,which might be achieved by regulating mTOR signaling pathways.
