摘要
从实验室保存的海带降解菌群中分离出3种不同菌落形态的菌株(Lj1、Lj2和Lj3),经交叉划线法和滤纸片法对分离得到的菌株进行相互作用分析,结果发现Lj1与Lj2之间存在相互促进作用;应用分离得到的菌株混合发酵海带,在235 nm波长处测定产物中海藻寡糖的含量,结果表明以Lj1与Lj2组合为最高,稀释50倍后的吸光度为0.784。经细菌16S rDNA测序鉴定,Lj1和Lj2分别为印度斯塔普氏菌和陶氏假单胞菌。采用Lj1与Lj2复合菌种发酵海带,3~4 d可使培养基中质量分数30%的湿海带的分解率达90%以上,产物中的海藻寡糖推测为单一组分的三糖。
Three strains with different colony morphologies(Lj1,Lj2 and Lj3)are isolated from the kelp-degrading flora preserved in the laboratory.The interaction analysis of the isolated strains by the cross-streak method and the filter paper method shows that there is a mutual promotion effect between Lj1 and Lj2;the separated strains are used for mixed fermentation of kelp,and the content of algae-oligosaccharides in the product is measured at a wavelength of 235 nm.The results show that the combination of Lj1 and Lj2 is the highest,and the absorbance after dilution 50 times is 0.784.After bacterial 16S rDNA sequencing,Lj1 and Lj2 are identified as Stappia indica and Pseudomonas taeanensis respectively.Using Lj1 and Lj2 composite strains to ferment kelp,the decomposition rate of wet kelp with a mass fraction of 30%in the culture medium can reach more than 90%in 3 to 4 days,and the algae-oligosaccharides in the product are presumed to be trisaccharide of a single component.
