金银花木樨草苷对大鼠抑郁及中枢海马神经递质调控的影响 被引量:3

摘要 目的研究金银花木樨草苷对大鼠抑郁及中枢海马神经递质调控的影响。方法将48只成年健康的SD大鼠随机分为空白组、模型组、氟西汀组和金银花木樨草苷高、中、低(100 mg/kg、50 mg/kg、25 mg/kg)组。采用慢性不可预知性应激(chronicunpredictable mild stress,CUMS)结合孤养方法构建抑郁模型,旷场实验、悬尾实验、强迫游泳实验等考察药物对抑郁大鼠行为学的影响,HPLC-F检测海马中5-HT等单胺递质水平,Western blot和RT-PCR测定海马组织BDNF蛋白和mRNA的表达。结果与空白组比较,模型组大鼠矿场实验自主活动量下降,悬尾不动时间和游泳不动时间延长(P<0.05);海马区神经递质5-HT、NE及DA水平降低(P<0.05),BDNF表达下调(P<0.05)。结论金银花木樨草苷可逆转并改善慢性应激引起的抑郁样行为,其可能的机制是上调海马BDNF表达,增加中枢单胺类神经递质水平。
