摘要
为了解我国两广地区猪非典型瘟病毒(APPV)的流行及变异情况,本研究于2017年~2018年从该地区的两个规模化养殖场采集了10份患先天性震颤仔猪的各组织病料样品,利用RT-PCR进行APPV检测,挑选3份阳性样品进行该病毒的开放阅读框(ORF)基因序列的RT-PCR扩增、测序,并对ORF基因、该基因包含的E2和P7基因进行核苷酸和氨基酸序列同源性分析及该3种基因的遗传进化分析。结果显示,10份病料样品均为APPV阳性,采用RT-PCR分段扩增获得3株APPV的ORF基因全长序列。ORF基因序列分析显示,3株APPV之间的核苷酸同源性为81.6%~99.8%,与41株中国株的同源性为80.8%~99.3%,与19株国外参考株的同源性为81.3%~91.1%;E2和P7基因的同源性分析结果与ORF基因同源性分析结果一致。ORF、E2和P7基因的遗传进化树显示,广西猪场分离的GX-GL1株和GX-GL67株属于基因1型,而广东猪场分离的GD-CT4株属于基因3型。上述结果表明3株APPV与中国株的亲缘关系更近,两广地区的APPV基因型呈现地域差异性,本研究有助于了解APPV在中国的流行情况。
To investigate the prevalence and variation of atypical porcine pestivirus(APPV)in Guangxi and Guangdong provinces,the tissue samples of 10 piglets with congenital tremors were collected from two farms during 2017 to 2018,and APPV was detected by reverse transcription-PCR(RT-PCR).The open reading frame(ORF)gene of three positive samples was amplified and sequenced,and homology analysis and phylogenetic analysis of ORF,P7 and E2 genes were performed.The results showed that 10 samples were APPV positive and ORF sequences of three APPV isolates were obtained.The ORF sequence analysis showed that three APPV isolates shared 81.6%-99.8%nucleotide identity among themselves,80.8%-99.3%with 41 isolates from China,and 81.3%-91.1%with 19 isolates from other countries.The phylogenetic analysis of ORF,P7 and E2 genes showed that GX-GL1 and GX-GL67 isolates from Guangxi farms belong to genotype 1,while GD-CT4 from Guangdong farms belongs to genotype 3.The three APPV isolates displayed the closer relationship to China isolates,the genotype of APPV isolates in Guangdong and Guangxi showed regional differences.This study provides the prevalence information of APPV in Guangxi and Guangdong provinces.
