玉米组蛋白乙酰转移酶的鉴定与表达规律分析 被引量:5

摘要 组蛋白乙酰转移酶(Histone acetyltransferases,HATs)催化组蛋白乙酰化,通过调节组蛋白乙酰化状态影响染色质的结构,进而影响基因的转录,在植物的生长、发育和胁迫反应过程中发挥重要的调控作用。目前有关玉米HATs家族基因的系统研究尚未见报道。本研究利用生物信息学方法,对玉米HATs家族基因进行系统进化分析、保守结构域分析、组织表达特性分析以及生物和非生物胁迫下的表达规律分析。结果发现,玉米HATs家族基因可以分为GANT、MYST和CBP 3个亚家族,均包含多个保守的结构域,在不同组织和生物/非生物胁迫过程中表达水平呈现明显的不同。利用Real-time PCR技术,检测HATs家族基因在激素处理玉米后的表达水平,发现该家族成员在茉莉酸和水杨酸处理后表现出不同的表达规律。这些结果表明玉米HATs家族基因在玉米生长发育以及响应生物和非生物胁迫过程发挥重要的作用,为阐明玉米HATs家族基因的功能及其调控机制奠定了基础。 Histone acetyltransferases(HATs)mediate histone acetylation and regulate the status of histone acetylation,which modifies chromatin structure,affects gene transcription and thus plays a crucial regulatory role in plant growth,development and stress responses.However,the systematic analysis of HATs family genes in maize has not been reported yet.In this study,bioinformatics methods were used to analyze the phylogenetic evolution,conserved domain,tissue expression characteristics and expression patterns of the HATs family genes in maize under biological and abiotic stress.It was found that the HATs family genes in maize can be divided into three subfamilies of GANT,MYST and CBP,which all contain multiple conserved domains,and their expression levels were significantly different in different tissues and under different biological/abiotic stress.Real-time PCR technology were used to detect the expression levels of the HATs family genes in maize after hormone treatment.It was found that the HATs family genes showed different expression patterns after jasmonic acid and salicylic acid treatment.These results indicate that the maize HATs family genes play an important role in the growth and development of maize and in response to biological and abiotic stress,which lays a foundation for elucidating the function and regulatory mechanism of maize HATs family genes.
