面部穴位埋线对变应性鼻炎大鼠模型紧密连接蛋白Claudin-1和Occludin表达的影响 被引量:7

Effects of catgut implantation at acupoint of face on expression of the Tight Junction Proteins Claudin-1 and Occludin of rats with allergic rhinitis

摘要 目的观察面部穴位埋线对变应性鼻炎大鼠模型鼻黏膜上皮紧密连接蛋白Claudin-1和Occludin表达的影响。方法 SD大鼠随机分成治疗组、模型组和空白组,每组6只。治疗组和模型组卵清蛋白造模。模型完成后,模型组和空白组不予干预,治疗组迎香穴(LI20)埋线,每周1次,连续2周。观察大鼠变应性鼻炎相关的行为学评分改变,HE染色后光镜下观察鼻黏膜炎性反应情况,电镜下观察鼻黏膜上皮细胞内质网和紧密连接,蛋白质印迹法测定Claudin-1和Occludin灰度值。结果与模型组相比,治疗组流涕、喷嚏和抓挠症状明显改善(P<0.05),鼻黏膜纤毛倒伏、脱落、嗜酸性粒细胞浸润等情况及上皮细胞内质网扩张、脱颗粒均缓解,上皮细胞紧密连接间隙减小。3组大鼠Claudin-1和Occludin灰度值经两两比较,差异均具有统计学意义(P<0.05)。结论面部穴位埋线可以上调紧密连接蛋白Claudin-1和Occludin的表达,增强黏膜屏障的功能,减少过敏原的入侵,这可能是穴位埋线治疗AR的机制之一。 Objective To observe the effect of catgut implantation at acupoint of face on the expression of claudin-1 and Occludin in the nasal mucosa of rat models of allergic rhinitis. Methods We divided SD rats into treatment group, model group and blank group randomly, with 6 rats in each group. Animal models of allergic rhinitis were made with ovalbumin in the treatment and model groups. After the model was completed, the treatment group were treated by catgut implantation at Yingxiang(LI20), once a week for 2 weeks. Observe changes in behavioral scores related to allergic rhinitis in rats, observe nasal mucosa inflammation under light microscope after HE staining, observe endoplasmic reticulum and tight junction of nasal mucosa epithelium under electron microscope, and determine Claudin-1 and Claudin-1 by Western blotting Occludin gray value. Results The symptoms of runny nose, sneezing, and scratching were significantly improved in the treatment group, nasal mucosal ciliary lodging, shedding, eosinophil infiltration, and endothelial reticulum expansion and degranulation of epithelial cells were all relieved compared with the model group. The tight junction gap of nasal mucosa epithelium in the treatment group was smaller than that in the model group.The gray values of Claudin-1 and Occludin in the three groups of rats were compared in pairs, and the differences were statistically significant(P <0.01). Conclusion Catgut implantation at acupoint of face can up-regulate the expression of tight junction proteins Claudin-1 and Occludin, and enhance the function of the nasal mucosal barrier, reduce allergen invasion. This may be one of the mechanisms of catgut implantation at acupoint for the treatment of AR.
