摘要
目的:研究大米草黄酮(DMC-fla)对人食管癌EC9706细胞侵袭力的影响和潜在作用机制。方法:通过细胞划痕实验和Transwell小室穿透实验评估大米草黄酮对EC9706细胞侵袭迁移能力的影响;利用RNA-seq和MicroRNA测序技术揭示大米草黄酮对EC9706细胞mRNA和miRNAs表达谱的影响和筛选关键性差异表达靶基因;Real-time qPCR验证所筛选出的靶基因的差异化表达。结果:在大米草黄酮作用下,EC9706细胞的划痕愈合能力和Mareigel胶穿透力都显著性下降;RNA-seq和MicroRNA测序筛选出3个与食管癌细胞侵袭力正相关但在大米草黄酮作用下显著差异性表达的靶基因位点;qPCR验证了整合素β6(ITGB6)、纤溶酶原激活物抑制因子1(SERPINE1)和miR-205这3个靶基因表达受大米草黄酮作用下调。结论:大米草黄酮通过下调ITGB6、SERPINE1和miR-205的表达抑制人食管癌EC9706细胞的侵袭力。
Objective:To evaluate the effect of flavonoid extract from Spartinaanglica(DMC-fla)on invasion capacity of esophageal cancer cell line EC9706 and its potential mechanism.Methods:Cell wound scratch assay and transwell chamber assay were applied to evaluate the invasion capacity of EC9706 cells under DMC-fla treatment;RNA-seq and Micro RNA sequencing were used to screen the target gene sites related with DMC-fla treatment;the screened target gene sites were verified by real-time qPCR.Results:The invasion capacity of EC9706 cells was dramatically reduced by DMC-fla treatment;3 gene sites were selected as target sites for DMC-fla associated with the invasion capacity of EC9706 cells;qPCR verified integrinβ6(ITGB6),plasminogen activator inhibitor1(SERPINE1)and miR-205 were down-regulated by DMC-fla.Conclusion:The flavonoid of Spartinaanglica inhibited the invasion capacity of esophageal carcinoma EC9706 cells via ITGB6,SERPINE1 and miR-205 down-regulation.
