四妙散治疗肾病综合征、痛风及湿疹的系统生物学研究 被引量:10

A Systematic Biology Study of Simiao Decoction(四妙散)for Treating Different Diseases with the Same Method on Nephrotic Syndrome,Gout,and Eczema

摘要 目的:运用系统生物学方法探究四妙散的药效物质基础与作用靶点,探讨其异病同治治疗肾病综合征(简称肾病)、痛风及湿疹的机制。方法:根据化合物药代动力学性质,筛选四妙散活性成分并预测其靶点;取四妙散潜在靶点与疾病靶点的交集,即治疗靶点;用STRING行靶点蛋白质相互作用(PPI)分析;用Cytoscape构建和分析网络;用GlueGO行基因本体(GO)及基因组百科全书(KEGG)功能富集分析。结果:筛选出47个四妙散活性成分,其中33个成分可通过47个靶点治疗肾病、痛风及湿疹;3种疾病有6个共同靶点,其中RELA、FOS、IL6和CASP3在3种疾病中均有重要地位。槲皮素、汉黄芩素、β-谷甾醇及豆甾醇是四妙散的核心药效物质,它们主要通过抑制炎症、免疫紊乱、氧化应激及凋亡实现异病同治,主要涉及IL-17、TNF、HIF-1及凋亡通路。结论:运用系统生物学方法探究四妙散的多维药理作用,阐释其异病同治治疗肾病、痛风及湿疹的分子机制,可为其临床应用和进一步科学研究提供思路。 Objective: To explore the medicinal substances and potential targets of Simiao Decoction(SMD based on systematic biology, then to elucidate the mechanisms of treating different diseases with the same method on nephrotic syndrome(NS), gout, and eczema. Methods: The bioactive compounds of SMD were screened according to their pharmacokinetic properties, and the potential targets of SMD was predicted. The intersection of potential targets of SMD and diseases targets were regarded as the targets of SMD in treatment of three diseases. Target protein-protein interaction(PPI) analysis was performed by STRING. The networks were constructed and analyzed using Cytoscape. Gene ontology(GO) and Kyoto encyclopedia of genes and genomes(KEGG) functional enrichment analyses were conducted using Glue GO. Results: In this study, 47 bioactive compounds of SMD were screened out, among which 33 might treat NS, gout and eczema through 47 targets. The three diseases had six common targets, in addition, RELA, FOS, IL6 and CASP3 played important roles in the promotion of three diseases. Quercetin, wogonin, β-sitosterol and stigmasterol were the main pharmacodynamic substances of SMD, which might attenuate inflammation, immune disorder, oxidative stress and apoptosis in treatment of three diseases, through IL-17, TNF, HIF-1 and apoptosis involved pathways. Conclusions: In this study, a network pharmacology approach was used to explore the multidimensional pharmacological effects of SMD, elucidating its molecular mechanisms of treating different diseases with the same method on NS, gout and eczema,which might provide new insights for the clinical practice and further research of SMD.
