摘要
目的基于网络药理学方法探讨宣痹汤治疗痛风的作用机制。方法运用TCMSP、BATMAN-TCM数据库筛选宣痹汤活性成分和对应靶点,借助Genecard、OMIM及GenCLiP 3数据库收集痛风的作用靶点并通过venny 2.1软件筛选出药物-疾病共同作用靶点。随后通过CytoNCA进行拓扑网络分析刷选出重要靶点,然后利用BINGO和MCODE插件对重要靶基因进行GO功能富集分析和聚类分析,最后再利用ClueGO插件进行KEGG通路分析并与CTD数据库中痛风相关通路进行匹配分析。结果预测宣痹汤治疗痛风的有效成分44个及共同作用靶点82个,获得IL1B、PTGS2、IL6、MAPK1、IL2、IL10等核心靶点32个,GO富集分析显示其干预炎症、免疫、核酸代谢等生物学过程,KEGG通路分析显示IL-17信号通路、TNF信号通路、NF-κB信号通路、NOD样受体信号通路、Toll样受体信号通路等为显著性较高的通路。结论宣痹汤可通过抑制炎症因子及相关的炎症通路,调控核酸代谢的过程,进而抑制痛风中的炎症反应,减少尿酸生成和排泄,从而发挥抗痛风的作用。此外,宣痹汤对于痛风合并心血管疾病、类风湿性关节炎可能存在潜在的治疗作用。
Objective To study the therapeutic mechanism of Xuanbi decoction in the treatment of gout based on network pharmacology.Methods Based on TCMSP and BATMAN-TCM databases,the active compounds of Xuanbi decoction were screened.The targets of gout were collected by means of the Genecards and OMIM and GenCLiP 3 databases and the drug-disease targets were screened by venny2.1 software.Subsequently,network related to gout was constructed by using Cytoscape(3.7.2)software and CytoNCA were applied to analyze the network.Then,GO function analysis and clustering analysis of target genes were conducted by using BINGO and MCODE plug-ins.Finally,KEGG pathway enrichment analysis were performed using ClueGO,and matched with gout-related pathways in the CTD database.Results Forty-four active ingredients and 82 targets were predicted in Xuanbi decoction for treating gout.Thirty-two key genes were obtained.GO analysis results showed that the biological function of key genes mainly involved in inflammation,immunity,and nucleic acid metabolism.KEGG pathway analysis showed that the significant pathways were IL-17 signaling pathway,TNF signaling pathway,NF-κB signaling pathway,NOD-like receptor signaling pathway,Toll-like receptor signaling pathways,etc.Conclusion Xuanbi decoction suppresses the inflammatory response in gout and reduces the production and excretion of uric acid by inhibiting inflammatory factors and inflammatory pathways,and regulates the process of nucleic acid metabolism,thereby exert an anti-gout effect.In addition,there may be a potential effect of Xuanbi decoction on cardiovascular disease and rheumatoid arthritis complications with gout.
