活血润肠软胶囊挥发油的提取工艺优化及气相色谱质谱分析 被引量:4

Optimization of the Extraction Process and GC-MS Analysis of Volatile Oil from Huoxue Runchang Soft Capsules

摘要 目的探讨水蒸气蒸馏法提取活血润肠软胶囊中当归、羌活和防风中挥发油的最佳提取工艺条件。方法采用正交试验法,以提取率为评价指标,考察加水量、浸泡时间、提取时间和加盐量对挥发油提取率的影响,采用气相色谱质谱(GC-MS)法分析其挥发油的化学成分。结果最佳提取工艺条件为加6倍量水,浸泡1.5 h,提取4 h,此条件下挥发油的提取率为0.34%;GC-MS法分析得到的主要成分为1,4-双(1-甲基乙基)-苯、1 R-α-蒎烯、β-蒎烯、醋酸呋喃酯、(R)-4-甲基-1-(1-甲基乙基)-3-环己烯-1-醇、柠檬烯等。结论浸泡时间、加水量和提取时间对挥发油的提取率影响不显著,影响顺序为浸泡时间>加水量>提取时间。 Objective To explore the optimal extraction process of volatile oil in Angelica sinensis,Notopterygium incisum,and Radix saposhnikoviae from Huoxue Runchang Soft Capsules by steam distillation.Methods Orthogonal experimental design was used to evaluate the extraction rate.The influence of the amount of water,soaking time,extraction time and the amount of salt added on the extraction rate of volatile oil was investigated.The chemical composition of the volatile oil was analyzed by gas chromatography-mass spectrometry( GC-MS).Results The optimal extraction process was adding 6 times the amount of water,soaking for 1.5 h,and extracting for 4 h.Under these conditions,the extraction rate of the volatile oil was 0.34%;the main component obtained by GC-MS analysis was 1,4-double( 1-methylethyl)-benzene,1 R-α-pinene,β-pinene,furan acetate,( R)-4-methyl-1-( 1-methylethyl)-3-cyclohexene-1-alcohol,limonene,etc.Conclusion Soaking time,water addition and extraction time have no significant influence on the extraction rate of volatile oil.The order of influence is soaking time > water addition > extraction time.
