摘要
[目的]建立以银杏幼茎节为外植体的无菌接种体系。[方法]以银杏树当年生幼茎节为外植体,研究不同种类、不同浓度消毒剂、不同处理时间对外植体的消毒效果,及不同生长调节剂条件对愈伤的诱导。[结果]外植体最佳接种条件为用75%乙醇处理2 min后,10%次氯酸钠处理15 min,12%次氯酸钠处理10 min;0.8%新洁尔灭处理20 min;12%双氧水处理8~10 min,污染率和褐化率都相对较低。在0.3mol/L NAA和0.3 mol/L 6-BA组合,0.3 mol/L NAA和0.5 mol/L 6-BA组合中,银杏幼茎节的愈伤诱导率较高,愈伤的生长情况较好。[结论]该研究建立了以银杏幼茎节为外植体的再生体系,为其愈伤诱导及其进一步的生理研究提供了基础。
[Objective]To establish the tissue culture regeneration system of young stem nodes of Ginkgo biloba.[Method]Taking young stem nodes of annual G.biloba tree as explants,disinfection effect of different kinds,concentrations of disinfectants,treatment time on external implants,and different hormone conditions on callus induction were studied.[Result]The optimal inoculation conditions for explants were:after 75%alcohol treatment for 2 min,10%sodium hypochlorite treatment for 15 min and 12%sodium hypochlorite treatment for 10 min;0.8%bromogera mine for 20 min;12%hydrogen peroxide for 8-10 min;resulted in relatively low pollution rate and browning rate.In the hormone combination of 0.3 mol/L NAA and 0.3 mol/L 6-BA,0.3 mol/L NAA and 0.5 mol/L 6-BA,the callus induction rate of young stem nodes of G.biloba was higher and the callus growth was better.[Conclusion]This study established tissue culture regeneration system of G.biloba,which provided basis for its callus induction and further physiological studies.
