不同南药多糖对鸡传染性法氏囊病毒VP2蛋白免疫原性的影响 被引量:6

Effect of different traditional south Chinese medicine polysaccharide on immunogenicity of chicken infectious Bursal disease virus VP2 protein

摘要 为了研究不同南药多糖对鸡传染性法氏囊病毒(IBDV)VP2蛋白免疫原性的影响,试验采取水提醇沉法提取槟榔、巴戟天、砂仁、黄秋葵、牛大力、益智仁、广藿香多糖,对免疫增强剂的安全性进行评估,再将100只雏鸡随机分成10组,每组10只,分别为益智仁组、广藿香组、牛大力组、巴戟天组、黄秋葵组、槟榔组、砂仁组、VP2蛋白组、油乳组、空白对照组,将各多糖配合鸡IBDV VP2蛋白对雏鸡进行免疫,VP2蛋白组、油乳组、空白对照组分别免疫相同剂量的VP2蛋白、油乳免疫增强剂+VP2蛋白、生理盐水。各组雏鸡于7,14,21,28日龄空腹称重并运用ELISA法测定其血清IBDV特异性抗体水平,同时对雏鸡免疫器官指数的变化进行分析。结果表明:各组雏鸡的体重、体温及进食均正常,注射部位及全身未出现异常变化。14日龄牛大力组、砂仁组雏鸡体重极显著高于油乳组(P<0.01),且显著效果持续长久,而益智仁组、巴戟天组、槟榔组雏鸡则在二免后体重极显著高于油乳组(P<0.01)。巴戟天、黄秋葵、砂仁、槟榔均能提高雏鸡的法氏囊、脾脏免疫器官指数,其中砂仁多糖与槟榔多糖的增强效果优于其他南药多糖,21,28日龄砂仁组与槟榔组的IBDV抗体水平明显高于其他组,且砂仁组在峰值后下降趋势较其他组缓慢。砂仁多糖不仅能促进雏鸡生长发育,同时与IBDV VP2蛋白具有协同作用,能有效增强脾脏与法氏囊免疫功能,提高IBDV特异性抗体水平,显著提高疫苗效率。说明南药多糖对雏鸡的成长没有不良影响,具有较好的安全性。砂仁多糖具有较高的免疫增强效果,可以应用于鸡IBDV VP2蛋白疫苗的佐剂研究。 The main objective of this paper was to study the effect of different traditional south Chinese medicine polysaccharide on immunogenicity of chicken infectious bursal disease virus VP2 protein. Areca catechu polysaccharide, Morinda officinalis polysaccharide, Amomum longiligular polysaccharide,Abelmoschus esculentus(L.)Moench polysaccharide, Millettia speciosa Champ polysaccharide,Alpinia oxyphylla fructus polysaccharide and Pogostemon cablin polysaccharide were extract by water-extraction and alcohol-precipitation method and the safety of polysaccharide immune enhancer was evaluated. In the experiment, 100 chickens were randomly divided into 10 groups, 10 in each group, which were divided into Alpinia oxyphylla fructus group,Pogostemon cablin group,Millettia speciosa Champ group,Morinda officinalis group, Abelmoschus esculentus(L.)Moench group,Areca catechu group, Amomum longiligular group, VP2 protein group, oil emulsion group and blank control group. Each polysaccharide group was immunized with IBDV VP2 protein, and VP2 protein group, oil emulsion group and blank control group were immunized with the same agent quantity of VP2 protein, oil emulsion immunoenhancer + VP2 protein, normal saline, respectively. The chickens in each group were weighed on an empty stomach at the age of 7, 14, 21 and 28 days, and the specific antibody level of IBDV in serum was determined by ELISA. The changes of immune organ index were analyzed. The results showed that the body weight, body temperature and food intake of each group were normal, there was no abnormal change in the injection site and the whole body. The weight of 14-day old Millettia speciosa Champ group and Amomum longiligular group was significantly higher than that of the oil milk group(P<0.01), and the significant effect lasted for a long time, while the weight of Alpinia oxyphylla fructus group, Morinda officinalis group and Areca catechu group was significantly higher than that of the oil milk group after the second immunization(P<0.01).
