摘要
目的探讨新生儿巨细胞病毒(HCMV)感染与糖蛋白B基因(gB)分型及甘露糖结合凝集素(MBL)基因多态性相关性。方法选择2016年3月-2019年1月杭州市富阳区妇幼保健院收治的HCMV感染新生儿97例作为HCMV组,再随机抽取同期在医院分娩的健康新生儿60例作为对照组,检测HCMV感染新生儿HCMV gB基因分型以及两组新生儿MBL基因多态性并进行比较分析。结果97例HCMV感染新生儿中检出HCMV gB基因分型主要为gB1型(占45.36%)。两组新生儿-550位点G等位基因与C等位基因频率无明显差异,HCMV组新生儿-221位点G等位基因频率显著高于对照组(P<0.05),HCMV组新生儿+4位点T等位基因频率显著高于对照组(P<0.05)。HCMV组新生儿MBL外显子1+230位点G等位基因频率显著高于对照组(P<0.05)。HCMV组新生儿HYPA单体基因型频率显著低于对照组(P<0.05),两组新生儿LXPA、LYPB、LYPA、LYQA单体基因频率比较差异无统计学意义。结论新生儿巨细胞病毒感染主要以gB1型为优势基因型,其中部分患儿可出现两种HCMV病毒株感染。
OBJECTIVE To explore the correlation between neonatal human cytomegalovirus(HCMV)infection and glycoprotein B(gB)genotype and mannose-binding lectin(MBL)gene polymorphism.METHODS Totally 97 neonates with HCMV infection who were treated in Hangzhou Fuyang District Maternal and Child Health Hospital form Mar 2016 to Jan 2019 were assigned as the HCMV group,meanwhile,60 healthy neonates who were born in the hospital were randomly chosen as the control group.The HCMV gB genotypes of the neonates with HCMV infection were detected,and the MBL gene polymorphism was observed and compared between the two groups of neonates.RESULTS Among the HCMV gB genotypes that were detected in the 97 neonates with HCMV infection,45.36%were gB1.There were no significant differences in the frequencies of-550 locus G allele and C allele between the two groups of neonates(P>0.05),the frequency of-221 locus G allele of the HCMV group was significantly higher than that of the control group(P<0.05),the frequency of+4 locus T allele of the HCMV group was significantly higher than that of the control group(P<0.05).The frequency of G allele at exon 1+230 of MBL of the HCMV group was significantly higher than that of the control group(P<0.05).The frequency of HYPA genotype of the HCMV group was significantly lower than that of the control group(P<0.05);there were no significant differences in the frequencies of LXPA,LYPB,LYPA and LYQA genotypes between the two groups of neonates.CONCLUSION gB 1 is the predominant genotype among the neonates with HCMV infection,and some of the neonates may have the infections with two genotypes of HCMV.
