摘要
目的探究雷公藤内酯醇(triptolide,TPL)对脂多糖(lipopolysaccharide,LPS)诱导的足细胞损伤模型的保护作用。方法以LPS诱导的足细胞损伤模型为研究对象,设立空白对照组、药物组、模型组和实验组。采用免疫荧光法观察足细胞肌动蛋白细胞骨架的形态及粘着斑蛋白Vinculin的表达,激光共聚焦显微镜采集图像;Western blot检测各组血管生成素样蛋白3(angiopoietin-like protein 3,Angptl3)、CD2-相关蛋白(CD2-associated protein,CD2AP)、Synaptopodin以及Palladin的表达。结果TPL可以使足细胞细胞骨架排列趋于整齐,部分丝状结构完整,提高足细胞CD2AP(P<0.05)和Synaptopodin(P<0.001)、Palladin(P<0.05)及Vinculin的表达,同时可降低足细胞Angptl3的表达(P<0.05)。结论TPL可在一定程度上保护足细胞骨架,缓解LPS造成的细胞损伤。
Objective To explore the protective effect of triptolide(TPL)on lipopolysaccharide(LPS)-induced podocyte injury.Methods An LPS-induced podocyte injury model in vitro was adopted in the experiment.The podocytes were divided into 4 groups:blank control group,drug group,model group,and experimental group.We observed the rearrangement of the podocyte cytoskeleton and detected the expression of focal adhesion protein Vinculin by immunofluorescence.The expression of CD2-associated protein(CD2AP),synaptopodin,palladin,angiopoietin-like protein 3(Angptl3)were detected by Western Blot.Results In vitro LPS-induced podocyte injury,TPL intervention resulted in a relatively neat podocyte cytoskeletal arrangement,a partial intact filamentous structure,and increased expressions of CD2AP(P<0.05),Synaptopodin(P<0.001),Palladin(P<0.05)and Vinculin.Meanwhile,it significantly reversed LPS-induced upregulation of Angptl3(P<0.05).Conclusion TPL is effective for protecting the podocyte cytoskeleton to some extent and alleviating the cell damage caused by LPS stimulation.
