复方芪芎颗粒对关节炎大鼠滑膜组织Toll样受体4/髓样分化因子88途径的影响 被引量:4

Effects of Compound Qixiong Granules on Toll Like Receptor 4/Myeloid Differentiation Factor 88 Pathway in Synovial Tissue of Arthritis Rats

摘要 目的:评价不同剂量复方芪芎颗粒对Wistar大鼠关节炎的治疗作用,并探索其对Toll样受体4(Toll like receptor 4,TLR-4)/髓样分化因子88(myeloid differentiation factor 88,MyD88)途径的影响。方法:将SPF级健康雄性Wistar大鼠60只分为空白组、模型组、雷公藤多甙片组和复方芪芎颗粒低、中、高剂量组。除空白组10只大鼠外,其余50只大鼠用完全弗氏佐剂造模。造模成功且模型稳定后,开始灌胃给药。空白组及模型组每日给予生理盐水10 mL/kg,低、中、高剂量组每日分别给予0.03、0.04、0.05 g/mL复方芪芎颗粒溶液10 mL/kg,雷公藤多甙片组每日灌胃0.94 mg/kg的雷公藤多甙片水溶液。实验过程中定期观察大鼠足跖肿胀情况,并测量大鼠足跖厚度,连续治疗3周后,处死大鼠收集膝关节滑膜,检测滑膜组织中TLR4、MyD88蛋白和TLR4 mRNA的表达情况。结果:造模后各组大鼠足跖厚度明显增加,TLR4、MyD88蛋白与TLR4 mRNA表达水平升高(P<0.05)。治疗3周后各给药组大鼠较模型组足跖厚度有不同程度下降,TLR4、MyD88蛋白与TLR4 mRNA表达水平下降(P<0.05),其中复方芪芎颗粒中、高剂量组效果优于低剂量组与雷公藤多甙片组(P<0.05)。结论:复方芪芎颗粒可改善佐剂性关节炎大鼠关节炎症,且中、高剂量作用效果优于低剂量组与雷公藤多甙片组,其作用机制可能与抑制TLR4/MyD88信号通路有关。 Objective:To evaluate therapeutic effects of different doses of compound Qixiong granules in treating Wistar rats with arthritis,and explore its effects on toll like receptor 4(TLR4)/myeloid differentiation factor 8(MyD88)pathway.Methods:Sixty SPF healthy male Wistar rats were allocated to the blank group,the model group,tripterygium glycosides tablets group,low,moderate and high doses groups of compound Qixiong granules.Except ten rats in the blank group,other 50 rats were established into the models using complete freund's adjuvant.They began to take the medicine by intragastric administration after successfully modeling and the the models were stable.The blank group and the model group were given physiological saline each day,10 mL/kg,low,moderate and high doses groups were given with the solution of compound Qixiong granules in the concentations of 0.03,0.04 and 0.05 g/mL,10 mL/kg.To observe plantar swelling of the rats regularly in the experimental course,and detect plantar thickness of the rats,the rats were sacrificed after treating three consecutive weeks to collect the synovium of knee joint,and detect the expressions of TLR4,MyD88 protein and TLR4mRNA in synovial tissue.Results:After modeling,plantar thickness of the rats in different groups increased notably,the expressions of TLR4,MyD88 protein and TLR4mRNA lifted(P<0.05).After treating three weeks,plantar thickness of the rats lowered to different degrees in different medication groups compared with the model group,the expressions of TLR4,MyD88 protein and TLR4mRNA lowered(P<0.05),among them,moderate and high doses groups of compound Qixiong granules were superior to low dose group and tripterygium glycosides tablets group(P<0.05).Conclusion:Compound Qixiong granules could improve the arthritis of rats,moderate and high doses groups are superior to low dose group and tripterygium glycosides tablets group,and its mechanism might be related to inhibiting TLR4/MyD88 signaling pathway.
