摘要
目的研究葛根素对妊娠期糖尿病大鼠心肌损伤及心肌组织过氧化物酶体增殖物激活受体γ(PPARγ)表达的影响。方法将54只同期妊娠大鼠随机分为对照组、模型组与实验组,各18只。实验组与模型组大鼠通过腹腔注射链脲佐菌素构建妊娠期糖尿病大鼠模型,对照组注射等量生理盐水。建模成功后实验组大鼠灌胃葛根素160 mg·kg^-1,每天1次,模型组及对照组则灌胃等量生理盐水,连续干预2周。比较各组大鼠干预前后空腹血糖、心肌酶水平变化;以苏木精-伊红(HE)染色观察大鼠心肌组织病理学形态,以蛋白质印迹(WB)法检测心肌组织PPARγ蛋白表达情况。结果干预2周后,对照组、模型组及实验组大鼠血清肌酸激酶(CK)含量分别为(29.05±3.09),(40.46±3.11),(32.18±2.87)U·L^-1;血清肌酸激酶同工酶(CK-MB)含量分别为(39.56±3.16),(95.11±3.97),(60.21±2.99)U·L^-1;心肌组织PPARγ蛋白相对表达量分别为0.18±0.09,0.41±0.10,0.26±0.08。与干预前比,实验组大鼠干预1~2周后血清空腹血糖水平逐渐降低,干预2周后血清CK、CK-MB水平降低,且均低于同时间点模型组;实验组心肌组织PPARγ蛋白相对表达量较模型组降低,差异均有统计学意义(均P<0.05)。结论葛根素可有效恢复妊娠期糖尿病大鼠血糖水平,改善其心肌损伤,其作用机制可能与有效下调心肌组织PPARγ蛋白表达相关。
Objective To explore the effect of puerarin on the myocardial injury and myocardial tissue peroxisome proliferator activates receptorγ(PPARγ)expression of gestational diabetes rat.Methods Fifty-four pregnancy rats in the same period were randomly divided into control group,model group and test group,18 rats in each group.Rats in model group and test group were built gestational diabetes rat model by intraperitoneal injection of chain urea with cephalosporins,rats in control group were intervented by intraperitoneal injection of equal amount of normal saline.After the success of modeling,the rats in test group were treated with 160 mg·kg^-1 puerarin by gavage,1 time·d-1,rats in model group and control group were treated with equal amount of normal saline by gavage,continuous intervention for two weeks.The fasting plasma glucose,myocardial enzyme levels of rats before and after intervention were compared;the myocardial tissue pathology morphology of rats were observed by hematoxylin and eosin(HE)staining,the myocardial tissue PPARγprotein expression were detected by Western blot.Results Two weeks after intervention,the serum creatine kinase(CK)contents in control group,model group and test group were(29.05±3.09),(40.46±3.11),(32.18±2.87)U·L^-1;serum creatine kinase isoenzyme(CK-MB)contents were(39.56±3.16),(95.11±3.97),(60.21±2.99)U·L^-1;the relative expression of myocardial tissue PPARγprotein were 0.18±0.09,0.41±0.10,0.26±0.08.Compared with before intervention,the serum fasting plasma glucose level of rats in test group at 1 to 2 weeks after intervention were decreased gradually,serum CK,CK-MB levels at 2 weeks after intervention were decreased,which were lower than those in model group at the same time point;the the relative expression of myocardial tissue PPARγprotein in test group was lower than model group,the difference was statistically significant(all P<0.05).Conclusion Puerarin can restore the blood sugar levels of gestational diabetes rat effectively,improve the myocardial injury,the me
