丹参酮联合大黄素通过JAK/STAT3信号通路抑制结直肠癌细胞增殖、迁移的实验研究 被引量:3

摘要 目的:观察丹参酮联合大黄素通过JAK/STAT3信号通路抑制结直肠癌细胞的增殖、迁移。方法:实验分为四组,参照组(结直肠癌细胞株SW620),丹参酮组(结直肠癌细胞株+16μmol/L丹参酮)和大黄素组(结直肠癌细胞株+15μmol/L大黄素),联合组(结直肠癌细胞株+16μmol/L丹参酮+15μmol/L大黄素)。通过细胞转染、Western blot、细胞划痕实验等检测p-JAK和p-STAT3蛋白、细胞迁移、增殖、凋亡的表达。结果:丹参酮组和大黄素组对p-STAT3和p-JAK蛋白的抑制性均显著高于参照组(P均<0.05),丹参酮组和大黄素组的对p-STAT3和p-JAK蛋白的抑制性无明显差异(P>0.05),联合组对p-STAT3和p-JAK蛋白的抑制性高于丹参酮组和大黄素组(P<0.05);48 h后,联合组的增殖能力明显低于参照组(P<0.05),72 h后丹参酮组、大黄素组细胞增殖能力显著低于参照组(P<0.05),丹参酮组和大黄素组相比无明显差异(P<0.05),联合组明显低于丹参酮组和大黄素组(P<0.05),96 h后丹参酮组、大黄素组细胞增殖能力显著低于参照组(P<0.05),联合组明显低于丹参酮组和大黄素组(P<0.05);丹参酮组和大黄素组较参照组有明显的愈合(P<0.05),联合组细胞划痕愈合度较丹参酮组和大黄素组明显(P<0.05);丹参酮组和大黄素组较参照组促细胞凋亡能力明显增强(P<0.05),联合组较丹参酮组和大黄素促细胞凋亡能力显著增强(P<0.05)。结论:丹参酮联合大黄素通过JAK/STAT3信号通路,降低了SW620的增殖和迁移能力,促进了细胞凋亡,整体上遏制了结直肠癌的发展。 Objective:To study the inhibitory effect of tanshinone combined with emodin on the proliferation and migration of colorectal cancer cells through JAK/STAT3 signaling pathway.Methods:The experiment was divided into four groups:control group(SW620),tanshinone group(16μmol/L tanshinone)and emodin group(15μmol/L emodin)and combination group(16μmol/L tanshinone+15μmol/L emodin)through cell transfection,W.The expression of p-JAK and p-STAT3 protein,cell migration,proliferation and apoptosis were detected by estern blot and cell scratch test.Results:The inhibition of p-STAT3 and p-JAK protein in tanshinone group and emodin group was significantly higher than that in control group(P<0.05).There was no significant difference in the inhibition of p-STAT3 and p-JAK protein between tanshinone group and emodin group(P>0.05).The inhibition of p-STAT3 and p-JAK protein in combined group was higher than that in tanshinone group and emodin group(P<0.05).After 48 hours,the proliferation ability of the combined group was significantly lower than that of the control group(P<0.05),and began to show differences after 72 hours.The cell proliferation ability of the tanshinone group and emodin group was significantly lower than that of the control group(P<0.05),but there was no significant difference between the tanshinone group and emodin group(P<0.05),and the combined group was significantly lower than that of the tanshinone group and the emodin group(P<0.05).After 96 hours,the cell proliferation ability of tanshinone group and emodin group was significantly lower than that of the control group(P<0.05),the combined group was significantly lower than that of tanshinone group and emodin group(P<0.05),and the tanshinone group and emodin group healed better than that of the control group(P<0.05).The healing degree of scratches in combination group was more obvious than that in tanshinone group and emodin group(P<0.05);the apoptotic ability of tanshinone group and emodin group was significantly stronger than that in control group(P<0.05
