摘要
目的应用超声生物显微镜(UBM)观察肥胖小鼠心肌早期损伤情况,并探讨其与NLRP3炎症因子的关系。材料与方法选取120只6周龄雄性C57BL/6小鼠,喂以高脂饲料。选取符合要求的肥胖小鼠模型60只,随机分为肥胖模型组、肥胖+慢病毒空载体组、肥胖+NLRP3-siRNA慢病毒组,每组20只。采用UBM动态观察小鼠左心室超声生物学特性,包括形态学、血流动力学、功能、应变等的变化。检测NLRP3炎症小体蛋白水平变化。结果UBM显示对照组、肥胖组、肥胖+慢病毒空载体组、肥胖+NLRP3-siRNA组小鼠纵向应变组间差异均有统计学意义(P<0.05);肥胖组纵向应变值(7.35±1.37)、肥胖+慢病毒空载体组纵向应变值(11.06±1.22)均显著低于对照组(22.04±2.89)及肥胖+NLRP3-siRNA组(17.11±3.10),差异有统计学意义(P<0.05)。对照组和肥胖+NLRP3-siRNA组小鼠的NLRP3蛋白水平表达显著减低(P<0.05)。结论UBM可监测肥胖小鼠的心肌收缩功能改变情况,应变值可作为评估肥胖心肌收缩功能改变的一个准确和有效的指标,血清NLRP3炎症因子与肥胖心肌早期损伤有关。
Purpose To observe the early myocardial injury in obese mice by ultrasound biomicroscopy(UBM)and the relationship between the early myocardial injury and NLRP3 inflammatory factors in obese mice.Materials and Methods 1206-week male C57BL/6 mice were selected and fed with high-fat diet.Sixty obese mouse models that meet the requirements were selected and randomly divided into obesity model group,obesity+lentiviral empty vector group,obesity+NLRP3-siRNA lentivirus group,with 20 mice in each group.UBM was used to dynamically observe the ultrasound biological characteristics of the left ventricle of mice,including changes in morphology,hemodynamics,function and strain.The changes in NLRP3 inflammatory body protein levels were detected.Results UBM showed that there were statistically significant differences among the control group,obesity model group,obesity+lentivirus empty vector group and obesity+NLRP3-siRNA group in the longitudinal strain group(P<0.05).The longitudinal strain parameters of obesity group was 7.35±1.37 and that of obesity+lentivirus empty vector group was 11.06±1.22,which was significantly lower than those of the longitudinal strain parameters of control group 22.04±2.89 and the longitudinal strain parameters of obesity+NLRP3-siRNA group 17.11±3.10.The expression of NLRP3 protein in the control group and obesity+NLRP3-siRNA group decreased significantly.Conclusion UBM can monitor the changes of myocardial contractile function in obese mice,and the strain value can be used to evaluate the changes of myocardial contractile function in obese mice.
