摘要
目的探讨应用乙酰胆碱构建犬的急性房颤模型方法。方法 2018年1月至2019年6月,选择健康成年杂种犬20条,由郑州大学动物实验中心提供,犬龄1~2岁,体重(14.82±3.67) kg,持续静脉给予不同剂量乙酰胆碱,每个乙酰胆碱浓度给予3次短阵快速脉冲刺激,记录房颤诱发次数与持续时间。同时测量并记录不同乙酰胆碱剂量刺激部位的心房有效不应期。结果 20条犬成功构建具有不同房颤持续时间的急性房颤模型,实验动物心房刺激部位不应期随着乙酰胆碱剂量的增加而缩短[(118.31±7.68) ms比(77.38±8.21) ms,t=15.672,P<0.05]。实验犬达到最大房颤持续时间的乙酰胆碱平均浓度为(94.00±23.02) μg/(kg·min),最长平均房颤持续时间为(894.80±122.82) s。在一定乙酰胆碱剂量下房颤持续时间随着乙酰胆碱剂量的增加而增加,但是当乙酰胆碱剂量超过一定剂量时房颤持续时间反而随着乙酰胆碱剂量的增加而下降。结论应用阶梯计量的乙酰胆碱可以成功构建具有不同房颤持续时间的犬急性房颤模型。
Objective:To explore the establishment of a canine model of acute atrial fibrillation using acetylcholine.Methods:From January 2018 to June 2019,20 healthy adult hybrid dogs were selected and provided by animal experimental center of Zhengzhou University.The dogs were 1-2 years old and weighed[(14.82±3.67)kg].The different concentrations of acetylcholine were given.At each acetylcholine concentration,the subjects were given 3 bursts of stimulation,and the induction number and duration of atrial fibrillation were recorded.The effective atrial refractory period with different acetylcholine dose and stimulation sites was measured and recorded,simultaneously.Results:Totally,20 experimental canine models of acute atrial fibrillation were successfully established with different durations of atrial fibrillation,and the atrial refractory period in the stimulated sites was shortened along with the dose increase of acetylcholine[(118.31±7.68)ms vs.(77.38±8.21)ms,t=15.672,P<0.05].The average concentration of acetylcholine in experimental dogs reaching the maximum duration of atrial fibrillation was(94.00±23.02)μg/(kg·min),and the longest average duration of atrial fibrillation was(894.80±122.82)s.It was found that at a certain dose of acetylcholine,the duration of atrial fibrillation increased with the increase of acetylcholine dose,but when the acetylcholine dose exceeded a certain dose,the duration of atrial fibrillation decreased alternatively with the increase of acetylcholine dose,which revealed relationship between the duration of atrial fibrillation and the concentration of acetylcholine.Conclusion:The application of Ladder-type acetylcholine can successfully establish acute atrial fibrillation models in canine with different durations of atrial fibrillation.
