大鼠胸主动脉腹腔移植套管法的改良

摘要 目的:构建大鼠同种异体心脏移植物血管病模型,有效诱导心脏移植后的慢性排斥反应。方法:以Wistar大鼠为供体,将摘取的胸主动脉穿过自制的静脉留置针管后,翻转血管对称吻合、单侧打结固定,利用半针管将动脉套管植入到受体SD大鼠腹腔动脉中。术后7 d、28 d和56 d取移植动脉,常规行HE染色,观察血管形态变化,计算内膜厚度;免疫组织化学染色,观察α-平滑肌肌动蛋白(α-SMA)和增殖细胞核抗原(PCNA)相对表达量。结果:34只大鼠成功建模30只,成功率为88.24%;随时间增加,移植动脉不断被单核巨噬细胞侵蚀,弹性纤维断裂,内膜出现炎症细胞和平滑肌血细胞的堆积;与对照组相比,7 d、28 d和56 d各组动脉内膜厚度显著增加(均P<0.01),α-SMA表达量明显减少(均P<0.05),PCNA表达量显著增加(均P<0.05);且随着时间的增加,动脉内膜厚度不断增加,α-SMA的表达量逐渐减少,PCNA的表达量逐渐增加(均P<0.05)。结论:此种动脉套管的改良以及半针管的运用使动脉移植更加简便有效,可重复性高,内膜增厚显著,能成功诱导出移植后的慢性排斥反应。 Objective:To establish cardiac allograft vasculopathy model in rats,and induce chronic rejection after heart transplantation.Methods:Thoracic aorta obtained from the Wistar rat was passed through a self-made intravenous catheter.The inverted aorta was anastomosed symmetrically and fixed on one side.Then arterial cannula was implanted into the abdominal artery of Sprague-Dawley rats using semi-needle tube.The grafted artery was extracted on the 7 d,28 d and 56 d after transplantation.HE staining was performed routinely to observe the morphological changes of the artery and calculate the intimal thickness.The expression ofα-smooth muscle actin(α-SMA)and proliferating cell nuclear antigen(PCNA)were observed by immunohistochemistry.Results:There were 30 out of 34 models established,with a success rate of 88.24%.With the increase of time,the transplanted artery was continuously eroded by mononuclear macrophages,elastic fibers were fractured,and inflammatory cells and smooth muscle blood cells were accumulated in the intima.Compared with the control group,the intima thickness was obviously increased(all P<0.01),the expression ofα-SMA was obviously reduced(all P<0.05),and the expression of PCNA was markedly enhanced(all P<0.05)in each group at 7 d,28 d and 56 d.Moreover,with time going on,the intima thickness of the artery increased,the expression ofα-SMA decreased,and the expression of PCNA enhanced gradually(all P<0.05).Conclusion:The improvement of cannula and the application of semi-needle tube make the arterial transplantation simpler and more effective,with high repeatability and significant intimal thickening,which can successfully induce the chronic rejection after transplantation.
