血管紧张素Ⅱ及其受体在脂多糖诱导的大鼠急性肺、肾损伤中的变化 被引量:1

The changes of angiotensinⅡand its receptors in lipopolysaccharide-induced acute lung and kidney injury in rats

摘要 目的探讨血管紧张素Ⅱ(AngⅡ)及其受体在脂多糖(LPS)诱导的大鼠急性肺、肾损伤中的表达变化及意义。方法将Wistar大鼠48只随机分为对照组和LPS组,LPS组通过尾静脉注射LPS,对照组通过尾静脉注射等量的生理盐水,分别于2 h、6 h、12 h和24 h采集标本。通过HE染色观察肺、肾组织病理;测定肺湿/干重比、血清肌酐、尿素氮;应用放射免疫分析法检测血浆及肺、肾组织中AngⅡ浓度变化,Western blot、免疫组织化学法检测肺、肾组织血管紧张素受体1(AT1R)、血管紧张素受体2(AT2R)蛋白变化。结果与对照组相比,LPS组大鼠肺、肾组织病理显示不同程度损伤,肺湿/干重比、血清肌酐、尿素氮水平显著升高(P<0.05),血浆AngⅡ含量在2 h、6 h明显升高(P<0.05);肺、肾AngⅡ含量在各时间点均显著升高(P<0.05),肺、肾AT1R蛋白表达明显下降(P<0.05),AT2R蛋白表达明显升高(P<0.05),且AngⅡ、AT2R与肺、肾功能损伤指标呈正相关,AT1R与肺、肾功能损伤指标呈负相关。结论LPS导致大鼠的肺、肾功能出现损伤及肾素-血管紧张素系统改变,且AngⅡ及血管紧张素受体变化与肺、肾损伤存在相关性,血管紧张素Ⅱ及其受体可能参与内毒素诱导的肺、肾损伤。 Objective To investigate the expression and significance of the angiotensinⅡand its receptors in lipopolysaccharide(LPS)-induced acute lung injury and acute kidney injury in rats.Methods Forty eight Wistar rats were randomly divided into two groups:control group and endotoxin group(LPS group).LPS was injected through tail vein in LPS group,and the same amount of saline was injected through tail vein in control group.Samples were collected at 2 h,6 h,12 h and 24 h,respectively.The histopathology of lung and kidney was observed by HE staining.We detected lung wet/dry weight ratio,serum creatinine,urea nitrogen and AngⅡconcentration in plasma,lung and kidney tissues by radioimmunoassay.Western blot and immunohistochemistry were used to detect the expression changes of AT1R and AT2R in lung and kidney tissue.Results Compared with the control group,the pathology of lung and kidney tissue in LPS group showed different degrees of damage.The lung wet/dry weight ratio,serum creatinine and urea level in LPS group were significantly increased than that in control group(P<0.05).The AngⅡcontent in plasma increased significantly at 2 h and 6 h(P<0.05),and the expression level of AngⅡin lung and kidney increased significantly at all time points(P<0.05).The expression of AT1R in lung and kidney decreased significantly(P<0.05),while the AT2R protein expression increased significantly(P<0.05).Additionally the correlation analysis showed that the expression level of AngⅡand AT2R were positively correlated with lung and renal function,while the expression of AT1R was negatively correlated with lung and renal function.Conclusion LPS results in the damage of lung and kidney function and the change of renin-angiotensin system.The changes of AngⅡand angiotension receptors were correlated with lung and kidney injury.AngⅡand angiotension receptors may be involved in LPS induced lung and kidney injury.
