摘要
目的探讨芹菜素对2型糖尿病大鼠胫骨和腰椎骨密度的影响。方法50只大鼠随机选10只为空白组,40只大鼠经高糖高脂饮食喂养4周后,给予链脲佐菌素(40 mg/kg)一次性腹腔注射建立2型糖尿病大鼠模型,从中挑选造模成功30只大鼠,按体质量随机分为模型组、芹菜素组和骨化三醇组,分别以等量羧甲基纤维素钠、芹菜素[50 mg/(kg·d)]、骨化三醇[0.045μg/(kg·d)]连续灌胃8周,每两周测定一次体质量和空腹血糖。实验结束后,采用酶联免疫反应方法检测各组血清胰岛素(INS)、骨代谢转换指标[Ⅰ型胶原C端肽(CTX-Ⅰ)、骨钙素(OCN)],用双能X线骨密度仪检测胫骨和第2~5腰椎骨骨密度。结果与对照组相比,模型组空腹血糖高,血清INS、CTX-Ⅰ水平高,OCN水平低,腰椎和胫骨骨密度低(P均<0.05)。在给药第6周和第8周时,芹菜素组、骨化三醇组大鼠空腹血糖均低于模型组(P均<0.05)。与模型组相比,芹菜素组、骨化三醇组大鼠血清INS、CTX-Ⅰ水平低,OCN水平高(P均<0.05)。与模型组相比,芹菜素组、骨化三醇组腰椎和胫骨骨密度水平高(均P<0.05)。结论芹菜素能够降低实验性2型糖尿病大鼠血糖水平,增加INS分泌,提高骨密度水平,对2型糖尿病大鼠骨组织具有保护作用。
Objective To investigate the effect of apigenin(AP)on the bone density of tibia and lumbar vertebrae in rats with type 2 diabetes mellitus(T2DM).Methods Ten of 50 rats were randomly selected as the control group,and the remaining 40 rats were given intraperitoneal injection of streptozotocin(40 mg/kg)to establish the T2DM models after 4 weeks of high-sugar and high-fat diet.Thirty successfully modeled rats were selected.According to body weight,they were randomly divided into the model group(T2DM),apigenin group(T2DM+AP)and calcitriol group(T2DM+Cal).The above groups were given the same amount of sodium carboxymethyl cellulose,AP[50 mg/(kg·d)]and calcitriol[0.045 ug/(kg·d)]for 8 weeks,respectively,and the body weight and fasting glucose were measured every 2 weeks.At the end of the experiment,insulin,serum insulin(INS)and bone metabolism conversion indexes[C-terminal peptide of type I collagen(CTX-I)and osteocalcin(OCN)]were detected by ELISA in each group,and the bone density of tibia and the 2nd-5th lumbar vertebrae was measured by dual energy X-ray absorptiometry.Results Compared with the control group,the T2DM group had higher fasting blood glucose,serum INS,and CTX-I levels,lower OCN levels,and lower lumbar and tibial bone density(all P<0.05).At the 6th and 8th weeks of administration,the fasting blood glucose of rats in the T2DM+AP group and T2DM+Cal group was lower than that of the T2DM group(all P<0.05).Compared with the T2DM group,the levels of INS and CTX-I in the T2DM+AP group and the T2DM+Cal group were lower and the levels of OCN were higher(all P<0.05).Compared with the T2DM group,the T2DM+AP group,T2DM+Cal group and tibia group had higher BMD(all P<0.05).Conclusion Apigenin can reduce the blood glucose level,increase INS secretion,and improve bone density in the experimental T2DM rats,and has a protective effect on the bone tissues.
