摘要
目的:观察麦粒灸对胃热证胃黏膜损伤大鼠胃黏膜屏障的影响。方法:按随机数字表法将24只SD大鼠分为3组,分别为:正常组、模型组、麦粒灸组。除正常组外,模型组和麦粒灸组用辣椒混悬液和乙醇溶液灌胃。造模后除正常组外,其余分别予艾灸干预7 d,分别观察各组大鼠的一般行为学、胃黏膜的组织病理学变化,采用实时荧光定量(QT-PCR)检测各组大鼠胃黏膜组织TFF2、肿瘤坏死因子(TNF-α),采用ELISA测定各组大鼠血清TFF2含量。结果:与正常组比较,模型组胃热证大鼠毛发发黄,唇、舌、趾、爪、耳颜色偏红以及喜动、暴躁、叫声嘶哑、小便色黄和大便干结量少等;胃黏膜组织病理学检查可见胃黏膜皱襞出现萎缩,上皮细胞出现脱落坏死,细胞间隙增宽,炎症细胞浸润。与模型组比较,麦粒灸组大鼠较模型组大鼠好转,外观颜色有所改善,大小便情况较正常;胃黏膜组织病学检查麦粒灸胃经穴组胃黏膜皱襞破损有所恢复,细胞结构及固有腺体基本正常,炎症浸润减轻。与正常组相比,模型组胃黏膜中的TFF2 mRNA表达显著降低(P<0.01),TNF-αmRNA表达则显著升高(P<0.01),在麦粒灸组中则均得到了显著逆转(P<0.01或P<0.05)。模型组大鼠的血清TFF2含量显著降低(P<0.01),在麦粒灸组中均显著逆转(P<0.01)。结论:麦粒灸胃经穴可以促进胃热证胃黏膜损伤大鼠胃黏膜损伤修复,在一定程度上证明“热证可灸”具有可行性,该作用可能与艾灸胃经穴可以调节胃热证胃黏膜损伤大鼠胃黏膜屏障的作用有关。
Objective:To observe the effect of kernel moxibustion on gastric mucosal barrier in rats with Stomach-heat syndrome.Methods:24 SD rats were randomly divided into the normal group,the model group and the kernel moxibustion group.Except the normal group,the model group and the kernel moxibustion group were gavaged with capsaicin and ethanol solution.After seven days of moxibustion intervention,the general behavior and gastric mucosa's histopathological changes were observed in each group.TFF2 and TNF-αin gastric mucosa was detected by QT-PCR technique and the serum content of TFF2 was measured by ELISA method.Results:Compared to the normal group,the rats in the model group with Stomach-heat syndrome had yellow hair,red lips,tongue,toes,paws and ears,as well as abnormal movement,irritability,hoarse voice,yellow urine and less dry stool.Histopathological examination of gastric mucosa revealed atrophy of gastric mucosa folds,exfoliation and necrosis of epithelial cells,widening of cell space,and infiltration of inflammatory cells.Compared to the model group,the general conditions in the kernel moxibustion group were better improved than those in the model group.Gastric mucosa histological examination showed that gastric mucosa fold damage were recovered,cell structure and natural glands were basically normal,inflammatory infiltration was reduced in the kernel moxibustion group.Compared to that in the normal group,the expression of TFF2 mRNA was significantly decreased(P<0.01),and the expression of TNF-αmRNA was significantly increased in the model group(P<0.01);which were both significantly improved in the kernel moxibustion group after the treatment(P<0.01,P<0.05).The serum content of TFF2 was significantly decreased in the model group(P<0.01),which was significantly reversed in the kernel moxibustion group(P<0.01).Conclusion:Kernel moxibustion can promote the repair of gastric mucosa injury in rats with Stomach-heat syndrome,which proves the feasibility of"heat syndrome being treated with moxibustion"to some exten
