山竹果皮总氧杂蒽酮通过调控肝癌扩增因子1表达抑制结直肠癌细胞增殖并诱导凋亡 被引量:2

摘要 目的探讨山竹果皮总氧杂蒽酮和肝癌扩增因子1(ALC1)对结直肠癌细胞增殖和凋亡的影响及作用机制。方法以不同浓度山竹果皮总氧杂蒽酮处理SW480细胞,MTT法检测细胞增殖。采用流式细胞仪、实时荧光定量聚合酶链反应(qRT-PCR)和蛋白质印迹法(Western blot)检测400μmol/L氧杂蒽酮对SW480细胞凋亡及细胞中B细胞淋巴瘤/白血病-2(Bcl-2)、Bcl-2相关X蛋白(BAX)和ALC1表达的影响。将ALC1 siRNA或pcDNA-ALC1转染至SW480细胞,MTT和流式细胞仪检测细胞增殖和凋亡情况。结果不同浓度山竹果皮总氧杂蒽酮均可有效抑制SW480细胞增殖(P﹤0.05),其增殖抑制率随总氧杂蒽酮浓度的增加而提高(P﹤0.05)。以400μmol/L氧杂蒽酮干预SW480细胞后,细胞凋亡率提高,细胞中Bcl-2和ALC1表达下调而BAX表达上调(P﹤0.05)。在SW480细胞中敲减ALC1,细胞增殖抑制率和凋亡率均明显升高(P﹤0.01);过表达ALC1可减弱总氧杂蒽酮对细胞的增殖抑制及凋亡促进作用(P﹤0.05)。结论山竹果皮总氧杂蒽酮能够调控ALC1表达,从而抑制SW480细胞增殖并诱导细胞凋亡。 Objective To investigate the effect of total xanthone in mangosteen peel and amplified in liver cancer 1(ALC1)on the proliferation and apoptosis of colorectal cancer cells and related underlying mechanism.Method SW480 cells were treated with different concentrations of total xanthone from mangosteen peel,and cell proliferation was detected by MTT assay.The apoptosis of SW480 cells and the expression of B cell lymphoma/leukemia-2(Bcl-2),B-cell lymphoma/leukemia-2 associated X protein(BAX)and ALC1 in the in vitro subjects after treatment with 400μmol/L oxanthrone were detected by flow cytometry,real-time quantitative polymerase chain reaction(qRT-PCR)and Western blot at different levels of expression,respectively.ALC1 siRNA or pcDNA-ALC1 were transfected into SW480 cells,and then cell proliferation and apoptosis were detected by MTT assay and flow cytometry respectively.Result Xanthone from the mangosteen peel with various concentrations could inhibit the proliferation of SW480 cells(P<0.05),and the proliferation inhibition rate increased with the uplifting of total xanthone concentration(P<0.05).After treating SW480 cells with 400μmol/L xanthone,both the apoptosis rate and the expression level of BAX were increased while the expression levels of Bcl-2 and ALC1 were decreased(P<0.01).Knockdown of ALC1 in target cells could inhibit proliferation and induce apoptosis of SW480 cells,and overexpression of ALC1 could attenuate the inhibition effect on cell proliferation and apoptosis by xanthone.Conclusion The total xanthone in mangosteen peel could inhibit the proliferation of SW480 cells and induce the apoptosis of related cells through regulating the expression of ALC1.
