以Al^3+为媒介的UV-Vis法测定多西环素

摘要 为建立一种间接测定多西环素含量的紫外—可见分光光度法,在pH=5.0的醋酸—醋酸钠缓冲溶液介质中,将多西环素与Al^3+进行络合反应,得到在392 nm处有明显的特征吸收的六元环配合物,通过测定该络合物的吸光度A来间接测定多西环素的含量。在7.50~22.50μg/mL的浓度范围内,该配合物的吸光度A与多西环素的浓度C呈现出良好的线性关系,A=0.0316C+0.0119,r=0.9997,检出限为1.8μg/mL。将方法用于实际样品的加标回收实验时,回收率为101.2%~104.9%,RSD为0.63%~1.2%。该方法简单准确、重复性好、精密度高,选择性好,可应用于多西环素制剂样品的定量分析。 An indirect UV-Vis spectrophotometry was established to determine doxycycline.In the acetic acid-sodium acetate buffer solution at pH 5.0,doxycycline could be complexed with Al^3+to form six-membered ring complexes,and the obvious characteristic absorption peak could be generated at 392 nm.And the absorbance of the complex could be measured to indirectly determine the content of doxycycline.In the concentration range of 7.50~22.50μg/mL,the absorbance of the complex presented a good linear relationship with the concentration of doxycycline,A=0.0316C+0.0119,r=0.9997.The detection limit was 1.8μg/mL.And the method was applied to actual samples,the recovery rates were between 101.2%and 104.9%,and relative standard deviations were between 0.63%and 1.2%.The method is simple,accurate,repeatable,precise and selective,and can be applied to the quantitative analysis of doxycycline.
