摘要
目的基于网络药理学探讨黄芪治疗慢性肾炎的作用靶点和机制。方法通过中药系统药理学数据库与分析平台(TCMSP)筛选出黄芪的有效中药活性成分及中药靶标,之后从GeneCards数据库筛出慢性肾炎的疾病靶标,然后利用Cytoscape3.7.0软件绘制得到相应调控网络,在String数据库中构建出黄芪与慢性肾炎共同靶标的蛋白相互作用网络。最后利用R软件对二者的共同靶标进行GO功能富集分析和KEGG通路富集分析。结果筛选后得到黄芪有效药物成分20个,对应的药物-疾病作用靶点共74个,包括VEGFA、蛋白激酶AKT1、CXCL8等。GO功能富集分析主要包括细胞因子受体结合、细胞因子活性、氧化还原酶活性等。涉及的KEGG通路主要包括HIF-1信号通路、NF-κB信号通路等。结论运用网络药理学研究方法可推测黄芪治疗慢性肾炎的主要活性成分、功能活动、作用机制和信号通路,为日后的进一步研究提供了理论支撑和思路。
Objective To explore the target and mechanism of astragalus in the treatment of chronic nephritis based on network pharmacology.Methods Through analysis platform(TCMSP)and database system pharmacology of Chinese medicine extract of astragalus effective active ingredient in traditional Chinese medicine and Chinese medicine targets,sieve out from GeneCards database only after the disease of chronic nephritis targets,and then draw Cytoscape3.7.0 software are used to get the corresponding regulation network,in the String database construct astragalus with chronic nephritis target protein interaction network.Finally,R software was used to carry out GO functional enrichment analysis and KEGG pathway enrichment analysis on the common targets of the two.Results After screening,20 effective drug components of Astragalus were obtained,and the corresponding drug-disease targets totaled 74,including VEGFA,protein kinase AKT1,CXCL8,etc.GO enrichment analysis mainly included cytokine receptor binding,cytokine activity,REDOX enzyme activity,etc.The KEGG pathways involved mainly include hypoxic cell stress-1(HIF-1)signaling pathway,NF-κB signaling pathway,etc.Conclusion The main active ingredients,functional activities,mechanism of action and signaling pathway of astragalus membranaceus can be predicted by using network pharmacology,which provides theoretical support and ideas for further study in the future.
