细粒棘球蚴病患者Tim-3/Galectin-9对Th17/Treg的影响作用 被引量:1

摘要 目的探讨人细粒棘球蚴感染中T细胞免疫球蛋白黏蛋白分子-3/半乳糖凝集素-9(Tim-3/Galectin-9)与Th17/Treg关系。方法收集28例细粒棘球蚴感染患者(CE组)以及30例健康体检人员(正常对照组)外周血,用酶联免疫吸附(ELISA)法检测细粒棘球蚴感染患者外周血白细胞介素-10(IL-10)、白细胞介素-17A(IL-17A)、转化生长因子β(TGF-β)的水平,实时定量PCR检测Tim-3 mRNA、Galectin-9 mRNA以及Th17转录因子(RORγt)和Treg细胞转录因子(FOXP3),并进行相关性分析。结果细粒棘球蚴感染患者CD4^+T细胞上Tim-3表达水平高于正常对照组,差异具有统计学意义(P<0.05);与正常对照组相比,CE组患者外周血中TGF-β、IL-10含量明显增加,差异具有统计学意义(P<0.05)。CE组患者血清中Galectin-9水平高于正常对照组,差异具有统计学意义(P<0.05)。而转录因子Foxp3在CE患者外周血单个核细胞(PBMC)中水平高于正常对照组,差异具有统计学意义(P<0.05)。Tim-3^+CD4^+T和Galectin-9分别与TGF-β和IL-10呈正相关(P<0.05)。Tim-3mRNA、Galectin-9 mRNA分别与FOXP3呈正相关(P<0.05)。结论Tim-3/Galectin-9参与了细粒棘球蚴感染患者的免疫应答,Tim-3/Galectin-9可能通过改变Treg细胞转录因子及细胞因子,促进Th17/Treg细胞平衡改变,导致细粒棘球蚴持续感染。 Objective To explore the relationship between Tim-3/Galectin-9 and Th17/Treg in cystic echinococcosis(CE)patients.Methods Peripheral blood of 28 CE patients and normal controls were collected.The levels of IL-10,IL-17A and TGF-βin the peripheral blood of CE patients were detected by ELISA.The mRNA expression of Tim-3,Galectin-9,Th17 transcription factors(RORγt)and Treg cell transcription factor(FOXP3)were detected by real-time quantitative PCR.Correlation analysis was performed.Results The expression level of Tim-3 on CD4^+T cells was higher than that in the normal control group,and the difference was statistically significant(P<0.05).Compared with the normal control group,the levels of TGF-βand IL-10 in peripheral blood of CE patients were significantly increased(P<0.05).The level of Galectin-9 in serum was higher in CE patients than the one in normal control group,and the difference was statistically significant(P<0.05).The level of FOXP3 in PBMC of CE patients was higher than that in normal control group,and the difference was statistically significant(P<0.05).Tim-3^+CD4^+T and Galectin-9 were positively correlated with the level of TGF-βand IL-10,respectively(P<0.05).The mRNA expression of Tim-3 and Galectin-9 were positively correlated with the one of FOXP3,respectively(P<0.05).Conclusion Tim-3/Galectin-9 is involved in the immune response of CE patients.Tim-3/Galectin-9 may promote the change of Th17/Treg cell balance by changing Treg cell transcription factors and cytokines,leading to sustained Echinococcus granulosus infection.
