沙利度胺对EAE小鼠的疗效观察及相关指标检测

摘要 为探讨沙利度胺对EAE的疗效及相关指标影响,以MOG35-55多肽序列为免疫原诱导C57BL/6小鼠EAE模型,同时设置正常对照组。在免疫诱导当天分别给予15和60 mg/kg沙利度胺治疗。对小鼠进行行为学评分至免疫诱导第20天,采集小鼠内眦静脉血并分离血清,ELISA检测IL-4、IL-6、TGF-β、IL-17、TNF-α水平;分离小鼠脾脏,FACS检测CD4^+CD25^+Foxp3^+Treg百分比。结果显示,对照组小鼠行为正常,无发病。沙利度胺治疗组小鼠行为学评分显著低于EAE模型组,且60 mg/kg给药组显著低于15 mg/kg给药组(P<0.05)。EAE组与正常对照组比较,Treg百分比明显降低(P<0.05);15 mg/kg沙利度胺给药组与EAE组比较,Treg百分比无显著变化;60 mg/kg沙利度胺给药组Treg百分比相较EAE组显著升高(P<0.05)。不同剂量沙利度胺给药组与EAE组比较,IL-4水平升高,IL-6、IL-17、TNF-α水平均降低,且60 mg/kg沙利度胺给药组变化更显著(均P<0.05);TGF-β在EAE组显著升高,且给药组与EAE组差异无统计学意义。由此,沙利度胺可升高EAE小鼠Treg百分比,抑制促炎因子分泌,有利于病情缓解。 We aimed to investigate the effect of thalidomide on EAE and to detect related indexes,MOG35-55 peptide sequence was used as immunogen in C57 BL/6 mice to develop EAE model.The EAE mice were divided into 3 groups,the first group was given 15 mg/kg thalidomide on the day of induction,the second group was given 60 mg/kg and the last group served as EAE control.The behavioral score of mice was monitored,and the serum was taken from the mice eyeball blood on the 20 th day of immune induction.The levels of IL-4,IL-6,TGF-β,IL-17 and TNF-αwere detected by ELISA.Mouse spleen was isolated and FACS was used to detect the proportion of CD4^+CD25^+Foxp3^+Treg.The results showed that mice in the normal control group showed normal behavior and no morbidity.The neurological function score of the drug treatment groups was significantly lower than that of the EAE model group,with the 60 mg/kg thalidomide group significantly lower than that of the 15 mg/kg group(P<0.05).The proportion of Treg was significantly higher in the EAE group than that in the control group(P<0.05).Compared with the EAE group,the proportion of Treg in 15 mg/kg thalidomide group did not show significant change,and the proportion of Treg in 60 mg/kg thalidomide group was higher than that of the EAE group(P<0.05).Compared with the EAE model group,the levels of IL-4 were increased in both 15 mg/kg and 60 mg/kg thalidomide groups,while the levels of IL-6,IL-17 and TNF-αwere decreased,with the change in the 60 mg/kg thalidomide group more significantly(P<0.05);the TGF-βlevel in the EAE group was increased,but not significantly compared with treatment group.In conclusion,thalidomide can reduce the percentage of Treg in EAE mice and inhibit the secretion of pro-inflammatory factors,which is beneficial to the remission of the disease.
