摘要
目的:探究复方苦参注射液、表柔比星及两药联合应用对人膀胱癌T24细胞增殖、凋亡的影响及其机制。方法:将膀胱癌T24细胞分为空白对照组、复方苦参注射液60μl/ml组、表柔比星4.6μmol/L组及联合给药(复方苦参注射液60μl/ml+表柔比星4.6μmol/L)组。CCK-8法观察膀胱癌T24细胞增殖,流式细胞术检测膀胱癌T24细胞凋亡,蛋白免疫印迹实验检测膀胱癌T24细胞凋亡关键蛋白的表达情况。结果:与空白对照组相比,复方苦参注射液60μl/ml和表柔比星4.6μmol/L能显著抑制膀胱癌T24细胞增殖(P<0.01),且联合用药能起到协同增效作用(P<0.01)。复方苦参注射液60μl/ml和表柔比星4.6μmol/L能显著诱导膀胱癌细胞凋亡(P<0.01),两药联合使用促进凋亡作用更加明显(P<0.01)。复方苦参注射液60μl/ml和表柔比星4.6μmol/L能下调膀胱癌细胞凋亡相关蛋白pro-Caspase 3和Bcl-2的表达(P<0.05),上调Cleaved PARP、Bax和Cleaved-Caspase 3蛋白的表达(P<0.05),降低Bcl-2/Bax比值(P<0.05),联合用药作用更加明显(P<0.05)。结论:复方苦参注射液与表柔比星能够抑制膀胱癌细胞增殖,促进膀胱癌细胞凋亡,两药联合应用起到协同增效作用。
Objective:To investigate the effect of compound kushen injection,epirubicin and their combination on the proliferation and apoptosis of human bladder cancer T24 cells and its mechanism.Methods:T24 cells were divided into the control group,60μl/ml compound kushen injection group,4.6μmol/L epirubicin group and the combination drug group(compound kushen 60μl/ml+epirubicin 4.6μmol/L).CCK-8 method was used to observe the proliferation of bladder cancer T24 cells,the flow cytometry was used to detect the apoptosis of bladder cancer T24 cells,Western blot was used to detect the expression of key proteins of bladder cancer T24 cells apoptosis.Results:Compared with the control group,60μl/ml compound kushen injection and 4.6μmol/L epirubicin significantly inhibited the proliferation of bladder cancer T24 cells(P<0.01),and the combination of drugs showed a synergistic effect(P<0.01).60μl/ml Compound kushen injection and 4.6μmol/L epirubicin significantly induced the apoptosis of bladder cancer cells(P<0.01),and the combination of the two drugs showed significant promote apoptosis(P<0.01).Compound kushen injection and epirubicin down regulated the expressions of Pro-caspase 3 and Bcl-2,up regulated the expressions of cleaved-PARP,Bax and cleaved-caspase 3,and down regulated the ratio of Bcl-2/Bax(P<0.05),and the effect of the combination drug was more significant(P<0.05).Conclusion:Compound kushen injection and epirubicin can inhibit the proliferation of bladder cancer cells and promote the apoptosis,and the combination of the two drugs has a synergistic effect.
