摘要
目的探讨补体因子H相关蛋白1基因表达与2型糖尿病发病机制的关系.方法将187例2型糖尿病患者设为观察组,200名健康体检者设为对照组,检测两组补体因子H相关蛋白1基因第5外显子rs12406079位点基因多态性.比较两组补体因子H相关蛋白1基因rs12406079位点基因型及等位基因,分析补体因子H相关蛋白1基因rs12406079位点基因型与糖尿病相关因素的关系.结果 补体因子H相关蛋白1基因第5外显子rs12406079位点基因型检测到GG型、AG型和AA型,观察组和对照组补体因子H相关蛋白1基因rs12406079位点基因型分布符合Hardy-Weinberg平衡.两组补体因子H相关蛋白1基因rs12406079位点基因型分布差异有统计学意义(P<0.01),观察组等位基因A比例显著高于对照组(P<0.01).补体因子H相关蛋白1基因rs12406079位点AG+AA型糖尿病患者胰岛素抵抗指数显著高于GG型患者(P<0.01).结论补体因子H相关蛋白1基因表达可能在糖尿病发病机制中有重要作用,其可能与患者胰岛素抵抗增强有关,值得进一步研究.
Objective To investigate the relationship between CFHR1 gene expression and pathogenesis of type 2 diabetes.Methods 187 patients with type 2 diabetes were selected as observation group,and 200 healthy people were selected as control group,the polymorphism of rs12406079 locus in exon 5 of CFHR1 gene was detected in both groups.The genotype and allele of the rs2406079 locus of the CFHR1 gene were compared between the two groups,and the relationship between the genotype of the rs12406079 locus of the CFHR1 gene and diabetes-related factors was analyzed.Results The genotype of rs12406079 in exon 5 of CFHR1 gene was detected as GG type,AG type and AA type,the genotype distribution of the rs12406079 locus of CFHR1 gene in the observation group and the control group conformed to Hardy-Weinberg equilibrium.The difference in genotype distribution of rs12406079 locus of CFHR1 gene between the two groups was statistically significant(P<10.01).The proportion of allele A in the observation group was significantly higher than that in the control group(P< 0.01).The insulin resistance index of patients with type AG+A A diabetes in CFHR1 gene r s l2406079 was significantly higher than that in patients with type GG(P<0.01).Conclusions The expression of CFHR1 gene may play an important role in the pathogenesis of diabetes,which may be related to the enhancement of insulin resistance in patients,and it is worthy of further study.
