网果酸模HPLC指纹图谱研究

摘要 目的建立网果酸模HPLC指纹图谱。方法采用Agilent SB-C18(4.6×250 mm,5μm)色谱柱,以甲醇-0.1%甲酸水溶液为流动相进行梯度洗脱,检测波长254 nm,流速1 m L/分钟,柱温25℃,进样量5μL,分析时间70分钟,以网果酸模样品指纹图谱共有峰的平均值作为其对照指纹图谱,采用相似度方法进行评价。结果建立了16批网果酸模样品的共有图谱,确定了10个共有峰,对6个峰进行了指认,16批样品与对照图谱相似度>91%;其中,"1号""2号""3号"地块网果酸模共有峰个数分别15、25、18,平均相似度分别为92.3%、97.4%、94.2%;1年生、2年生和3年生网果酸模共有峰个数分别25、15、14,平均相似度分别为98%、89.3%、95.8%;春、秋采集网果酸模共有峰个数分别22、10,平均相似度97.2%、94.0%;所建立的网果酸模指纹图谱测定方法精密度、稳定性和重复性良好。结论首次建立了网果酸模的HPLC特征指纹图谱,为网果酸模药材的鉴别、质量控制等提供了新的科学依据。 Objective To establish a high performance liquid chromatography(HPLC)fingerprint of Rumex chalepensis Mill..Methods Agilent SB-C18(4.6×250 mm,5μm)column was used for gradient elution using methanol-0.1%formic acid solution as mobile phase.The detection wavelength was 254 nm,the flow rate was 1 m L·min-1,the column temperature was 25℃,the sampling size was 5μL,and the analysis time was 70 minutes.The average value of fingerprint common peaks in Rumex chalepensis Mill.samples,was taken as the control fingerprint.And the method of similarity criteria was used for evaluation.Results The common fingerprints of 16 batches of Rumex chalepensis Mill.samples were established.10 common peaks were determined,and 6 peaks were identified.The similarity between the 16 batches of samples and the control fingerprint was more than 0.91%.Among them,the number of common peaks of Rumex chalepensis Mill.in‘No.1’,‘No.2’and‘No.3’plot are 15,25 and 18,respectively.And the average similarity was 92.3%,97.4%and 94.2%,respectively.The number of common peaks of annual,biennial and triennial Rumex chalepensis Mill.was 25,15 and 14,respectively.And the average similarity was98%,89.3%and 95.8%,respectively.The number of common peaks of Rumex chalepensis Mill.collected in spring and autumn was 22 and 10,respectively.And the average similarity was 97.2%and 94.0%,respectively.The established fingerprint method of Rumex chalepensis Mill.has characteristics of good precision,stability and repeatability.Conclusion The HPLC characteristic fingerprint of Rumex chalepensis Mill.was established for the first time,which provided a new scientific basis for the identification and quality control of Rumex chalepensis Mill..
