食源性单核细胞增生李斯特菌毒力岛4携带株的分型、毒力及生物被膜形成能力的测定 被引量:10

摘要 为了解食源性单核细胞增生李斯特菌(LM)毒力岛4(LIPI-4)携带株的血清分型、多位点序列分型、小鼠的半数致死量(LD50)和生物被膜的形成能力,本研究以新疆各师局2014年~2017年常规送检的1027份食品中分离的5株携带LIPI-4的LM(菌株编号LM461、LM873、LM928、LM2947、LM5573)为研究对象,采用PCR对分离菌株进行血清分型,对其7个看家基因的序列进行多位点序列分型(MLST),并通过腹腔注射法测定小鼠LD50,利用结晶紫染色法测定其生物被膜的形成能力。结果显示,5株食源性LM LIPI-4携带菌株血清型PCR分组均为1/2b、3b和7;多位点序列分析结果均为ST87(CC87);LM928、LM2947、LM5573、LM461和LM873菌株对小鼠的LD50分别为107.2 cfu、107.2 cfu、107.3 cfu、107.5 cfu、108.6 cfu;5株LM分离株均能形成生物被膜,其中LM928和LM2947形成能力最强,LM873最弱。本研究首次明确LIPI-4不仅仅存在于LM CC4克隆群,同时也存在于ST87型,CC87克隆群,该结论对LM LIPI-4的致病性及其致病机理的进一步探究具有重要意义。 The objective of this study is to determine the serotypes,multilocus sequence types,median lethal dose of mice and the ability of biofilm formation of food-borne Listeria monocytogenes(LM)carrying Listeria pathogenicity island 4(LIPI-4).In this study,5 strains of LM carrying LIPI-4(strain numbers:LM461,LM873,LM928,LM2947,LM5573)isolated from 1027 food samples submitted by various divisions in Xinjiang from 2014 to 2017 were used as research objects.PCR was used for serotyping,and the sequence of seven housekeeping genes was subjected to multilocus sequence typing(MLST).The median lethal doses(LD50)of mice were measured by intraperitoneal injection and the abilities of biofilm formation were measured by crystal violet staining method.The results showed that the serotype PCR groups of 5 strains of food-borne LM carrying LIPI-4 were 1/2b,3b and 7;the results of MLST analysis showed that these were all ST87(CC87).The LD50 of mice of LM928,LM2947,LM5573,LM461,and LM873 were 107.2 cfu,107.2 cfu,107.3 cfu,107.5 cfu and 108.6 cfu,respectively.All of them could form biofilms,among which LM928 and LM2947 had the strongest biofilm-forming ability and LM873 was the weakest.The results of this study are of great significance for further investigation of the pathogenicity and pathogenic mechanism of LM carrying LIPI-4.
