黄芪甲苷对尿酸钠诱导的大鼠急性痛风性关节炎的防治作用及炎性反应机制研究 被引量:20

Preventive and therapeutic effects of astragaloside IV on acute gouty arthritis induced by sodium urate in rats and the mechanism of inflammation

摘要 目的探讨黄芪甲苷对尿酸钠诱导的大鼠急性痛风性关节炎的防治作用及炎症反应机制研究。方法120只大鼠分为对照组、模型组、秋水仙碱组、黄芪甲苷低、中、高剂量组;对照组、模型组,给予腹腔注射0.9%氯化钠(3 ml/kg),持续7 d;秋水仙碱组、黄芪甲苷低、中、高剂量组给予腹腔注射相应药物,持续7 d;模型组、秋水仙碱组、黄芪甲苷各剂量组将制备的尿酸钠注射到右踝关节中诱导急性痛风。试验结束后,进行大鼠关节炎症积分、机械痛阈(PWT)及热刺激潜伏期(PWTL)的测定;同时测定膝关节滑膜组织中TLR2、NF-кB mRNA及蛋白水平以及IL-2、IL-6、TNF-α蛋白水平。结果与对照组比较,模型组PWT、PWTL评分降低,关节炎症积分、TLR2、NF-кB mRNA、IL-2、IL-6、TNF-α蛋白表达水平升高(P<0.05);与模型组比较,黄芪甲苷各剂量组PWT、PWTL评分升高,关节炎症积分、TLR2、NF-кB mRNA、蛋白表达水平,IL-2、IL-6、TNF-α蛋白表达水平降低(P<0.05),且随着黄芪甲苷给药剂量增加,PWT、PWTL评分逐渐升高,关节炎症积分、TLR2、NF-кB mRNA、IL-2、IL-6、TNF-α蛋白表达水平逐渐降低剂量-效应关系明显(P<0.05);与秋水仙碱组相比,黄芪甲苷低、中剂量组PWT、PWTL评分降低,关节炎症积分、TLR2、NF-кB mRNA、IL-2、IL-6、TNF-α蛋白表达水平升高(P<0.05);高剂量组PWT、PWTL评分略微降低,关节炎症积分、TLR2、NF-кB mRNA、IL-2、IL-6、TNF-α蛋白表达水平略微升高(P>0.05)。结论黄芪甲苷能明显抑制尿酸钠诱导的大鼠急性痛风性关节炎,其机制与黄芪甲苷能通过抑制炎症信号通路TLR2、NF-кB mRNA、蛋白表达水平的表达进而抑制IL-2、IL-6、TNF-α的表达有关。 Objective To investigate the preventive and therapeutic effects of astragaloside IV on acute gouty arthritis induced by sodium urate in rats and the mechanism of inflammation.Methods A total of 120 clean Wistar rats were divided into control group,model group,colchicine group,astragaloside IV low-dose group,astragaloside IV medium-dose group and astragaloside IV high-dose group.The rats in control group and model group were given intraperitoneal injection of 0.9%sodium chloride(3ml/kg)for 7d while the rats in other four groups were given intraperitoneal injection of corresponding drugs for 7d.The rats in control group were given injection of 0.9%sodium chloride into the right tibio-tarsal joint while those in other five groups were given injection of sodium urate at the same position to induce acute gout.After experiment,the joint inflammation score,pain thresholds including paw withdraw thermal latency(PWTL)and paw withdrawal threshold(PWT)were determined.At the same time,the levels of TLR2,NF-кB mRNA in knee joint synovium tissues,the levels of protein and the levels of IL-2,IL-6 and protein TNF were detected.Results As compared with those in control group,the scores of PWT and PWTL in model group were significantly decreased,however,the scores of the joint inflammation,the protein levels of TLR2 and NF-кB as well as the protein levels of IL-2,IL-6 and TNF-αwere significantly increased(P<0.05).With the increase of doses of astragaloside IV.the scores of PQPT and PWTL were significantly increased,but the scores of the joint inflammation,the levels of TLR2,NF-кB mRNA,the levels of protein and the levels of IL-2,IL-6,protein TNF were significantly decreased(P<0.05).As compared with those in colchicine group,the scores of PWT and PWTL in astragaloside IV low-dose group,astragaloside IV medium-dose group and astragaloside IV high-dose group were significantly decreased,however,the scores of joint inflammation,the protein levels of TLR2 and NF-кB as well as the protein levels of IL-2,IL-6,TNF-αwere significantl
