摘要
目的:探讨大鼠肝切除术后肝功能衰竭和肝再生机制。方法:SD大鼠随机分为假手术组、85%肝切除组、90%肝切除组。术后观察各组血清生化指标、肝实质损伤及肝再生相关指标、mTOR信号通路的变化。结果:90%肝切除诱导大鼠肝切除术后肝功能衰竭,与85%肝切除组相比,前者并未造成更加严重的实质损伤,但其肝再生程度受到明显抑制,并且mTOR信号通路活化水平较低。结论:mTOR信号通路介导的肝再生抑制可能是肝切除术后肝功能衰竭的病理生理机制。
Objective:To explore the effect of liver regeneration mediated by mTOR signal pathway in postoperative liver failure in rat.Methods:SD rats were randomly divided into sham operation(SO),85%partial hepatectomy(PH)and 90%PH.The changes of serum biochemical,hepatic parenchymal injury and regeneration,and the activity of mTOR singal pathway were recorded.Results:Compared with 85%hepatectomy,90%hepatectomy resulted in liver failure.No significant differences in liver parenchymal injury were detected between the two groups.The regeneration response were significantly lower,and the mTOR and its downstream effectors were not activated so obviously in 90%PH as those in 85%PH.Conclusion:mTOR-dependent suppression of remnant liver regeneration may lead to liver failure after liver resection.
