高原低氧环境下2型糖尿病患者血清HIF-1α、VEGF与尿液smad1表达的相关性分析 被引量:6

摘要 目的检测高原低氧环境下2型糖尿病患者血清HIF-1α、VEGF与尿液smad1水平,并分析其相关性。方法选取2016年1月-2018年1月本院确诊并治疗的T2DM患者82例为研究对象,选取同期健康体检者80例为对照组,选取对象居住地海拔均≥3 000m,采用酶联免疫吸附(ELISA)法检测血清HIF-1α、VEGF水平、尿液smad1水平,检测患者血压、甘油三酯(TG)、胆固醇(TC)、低密度脂蛋白胆固醇(LDL)、高密度脂蛋白胆固醇(HDL)水平、空腹血糖(FBG)。结果 2组年龄、性别、DBP、SBP、TG、TC差异无统计学意义(P>0.05),研究组BMI指数、血清HDL水平较对照组显著降低(P<0.05),LDL、FBG水平显著升高(P<0.05);研究组血清HIF-1α、VEGF及尿液smad1水平较对照组显著升高(P<0.05);血清HIF-1α与VEGF、尿液Smad1水平显著正相关(P<0.05),血清VEGF水平与尿液Smad1水平显著正相关(P<0.05);Logistic回归分析结果表明,FBG、HIF-1α、VEGF水平高是高原居民发生T2DM的独立危险因素(P<0.05)。结论高原低氧环境下T2DM患者血清HIF-1α、VEGF水平升高,是高原居民发生T2DM的危险因素,二者均与T2DM患者尿液Smad1水平正相关。 Objective To detect the levels of serum HIF-1α,VEGF and urinary Smad1 in type 2 diabetes mellitus patients under high altitude hypoxia environment,and to analyze their correlations.Methods 82 patients with T2 DM diagnosed and treated in our hospital from January 2016 to January 2018 were selected as the study subjects,and 80 healthy people in the same period were selected as the control group.The subjects′living areas were all above 3000 min altitude.The levels of serum HIF-1α,VEGF and urine Smad1 were detected by enzyme-linked immunosorbent assay(ELISA),and blood pressure,triglyceride(TG),cholesterol(TC),low density lipoprotein cholesterol(LDL),high density lipoprotein cholesterol(HDL)and fasting blood glucose(FBG)were measured.Results There was no significant difference in age,sex,DBP,SBP,TG and TC between the two groups(P>0.05).BMI index and serum HDL level in the study group were significantly lower than those in the control group(P<0.05),while LDL and FBG levels were significantly higher than those in the control group(P<0.05).The levels of serum HIF-1α,VEGF and urine Smad1 in the study group were significantly higher than those in the control group(P<0.05).Serum HIF-1αwas positively correlated with levels of VEGF and urine Smad1(P<0.05),and serum VEGF was positively correlated with urine Smad1(P<0.05).Logistic regression analysis showed that high levels of FBG,HIF-1αand VEGF were risk factors for T2 DM in plateau residents(P<0.05).Conclusion Levels of serum HIF-1αand VEGF in patients with T2 DM under high altitude hypoxia are elevated,which are risk factors for the occurrence of T2 DM in Plateau residents,Both of them are positively correlated with urine Smad1 level in patients with T2 DM.
