摘要
目的:构建β-氨基丙腈(BAPN)诱导的大鼠主动脉夹层模型,并探索最佳剂量和给药方式。方法:选用75只3周龄幼年SD大鼠,均购自中国科学院上海动物中心。采用随机数字表法将其平均分为A^E 5组进行实验,A组给予正常饲料,饮食量不限;B组正常饮食,给予生理盐水皮下注射;C组和D组分别给予混合了浓度分别为0.25%和0.40%的BAPN饲料喂养;E组正常饲料喂养,给予BAPN+生理盐水混合液皮下注射。实验为期6周,通过尸检的方法获取各组大鼠的主动脉标本,针对其中主动脉夹层标本进行形态学分析和病理学检测,并使用材料拉力仪分析标本的应力、应变及弹性模量等力学参数,采用 t检验比较各组间差异。 结果:C组中共11例出现主动脉夹层;D组中共3例出现主动脉夹层;E组中仅1例出现主动脉夹层。对照组(A组和B组)未发现主动脉夹层形成。E组大鼠的胸主动脉中膜厚度[(0.054±0.004) mm比(0.048±0.008) mm, t=2.598, P<0.05]和管壁面积[(0.620±0.074) mm 2比(0.557±0.030) mm 2, t=3.056, P<0.05]均明显高于对照组。 结论:口服低浓度(0.25%)BAPN能够高效构建大鼠AD模型,有助于主动脉夹层形成的力学机制研究。
Objective To construct a rat aortic dissection(AD)model byβ-aminopropionitrile(BAPN)and explore the optimal dosage and administration method.Methods In this experiment,753-week-old juvenile SD rats were selected and purchased from Shanghai Animal Center of Chinese Academy of Sciences.Random number table method was used to divide them into A-E 5 groups for experiment.Group A was given normal feed with unlimited dietary amount;Group B was given normal diet and was given subcutaneous injection of saline;Group C and D were given mixed concentrations of 0.25%and 0.40%BAPN feed was fed;group E was fed with normal feed and given a subcutaneous injection of BAPN+physiological saline mixture.The experiment lasted for 6 weeks.The aortic specimens of each group of rats were obtained by autopsy.Morphological analysis and pathological examination were performed on the aortic dissection specimens,and the stress,strain and elastic modulus of the specimens were analyzed using a material tension meter.For mechanical parameters,use t-test to compare the differences between groups.Results A total of 11 aortic dissections occurred in group C;3 in group D;1 in group E.The control group(group A and group B)did not find aortic dissection.The thickness of thoracic aorta intima in group E[(0.054±0.004)mm vs.(0.048±0.008)mm,t=2.598,P<0.05]and tube wall area[(0.620±0.074)mm2 vs.(0.557±0.030)mm2,t=3.056,P<0.05]were significantly higher than those in control group.Conclusion Oral BAPN at a low concentration(0.25%)can effectively build a rat AD model,and provide some help in the study of the mechanical mechanism of aortic dissection.
