摘要
目的:明确CXCL10/CXCR3轴与白癜风发病相关性,探索甘草酸可能参与免疫调节的过程。方法:选取18例健康对照者(健康对照组)、40例白癜风患者为研究对象。随机将40例白癜风患者分为对照组和试验组,每组各20例,对照组给予补肾活血方治疗,试验组给予补肾活血方联合复方甘草酸苷片治疗,两组均治疗2个月。治疗前,采用ELISA法检测健康对照者和白癜风患者CXCL10表达水平,采用流式法检测健康对照者和白癜风患者CXCR3^+CD4^+T细胞、CXCR3^+CD8^+T细胞表达频率;治疗后采用ELISA法检测白癜风患者和健康对照者外周血清中CXCL10表达水平,同时观察不同组间CXCL10表达差异。结果:与健康对照者比较,治疗前白癜风患者外周血清中CXCL10表达水平显著增高(P<0.001),治疗前白癜风患者CXCR3^+CD4^+在CD3^+细胞和CD4^+T细胞比值均显著升高(P<0.001),CXCR3^+CD8^+在CD3^+细胞和CD8^+T细胞比值亦显著升高(P<0.001)。与入组时比较,2个月后健康对照者外周血清中CXCL10表达水平无明显变化(P=0.442);治疗后,白癜风患者外周血清中CXCL10表达水平显著降低(P<0.01),但仍明显高于2个月后复测健康对照者(P<0.001);与对照组比较,试验组白癜风患者外周血清中CXCL10表达水平下降更明显(P<0.05)。结论:CXCL10/CXCR3轴与白癜风发病相关,甘草酸可能通过抑制CXCL10表达参与免疫调节过程。
Objective:To clarify the correlation between the CXCL10/CXCR3 axis and the incidence of vitiligo,and to explore the possible involvement of glycyrrhizin in the immune regulation process.Methods:18 healthy controls(healthy control group) and 40 patients with vitiligo were selected as the research subjects.Forty patients with vitiligo were randomly divided into a control group and a test group,with 20 cases in each group.The control group was treated with Bushen Huoxue Formular,and the test group was treated with Bushen Huoxue Formular combined with compound glycyrrhizin tablets.Both groups were treated for 2 months.Before treatment,ELISA method was used to detect the expression level of CXCL10 in healthy controls and vitiligo patients.Flow cytometry was used to detect the expression frequency of CXCR3^+CD4^+T cells and CXCR3^+CD8^+T cells in healthy controls and vitiligo patients;ELISA method was used after treatment The expression levels of CXCL10 in peripheral serum of patients with vitiligo and healthy controls,and observe the difference of CXCL10 expression between different groups.Results:Compared with healthy controls,the expression of CXCL10 in peripheral serum of patients with vitiligo before treatment was significantly increased(P<0.001),and the ratio of CXCR3^+CD4^+ in CD3^+ cells and CD4^+T cells in patients with vitiligo before treatment was significantly increased(P<0.001),CXCR3^+CD8^+ also increased significantly in the ratio of CD3^+ cells to CD8^+ T cells(P<0.001).Compared with the time of enrollment,the expression level of CXCL10 in the peripheral serum of healthy controls did not change significantly after 2 months(P=0.442);after treatment,the expression level of CXCL10 in the peripheral serum of patients with vitiligo decreased significantly(P<0.01),but it was still significant It was higher than that of healthy controls who were retested after 2 months(P<0.001);compared with the control group,the expression level of CXCL10 in the peripheral serum of vitiligo patients in the test group decreased
