摘要
目的探讨m iR-140-5p靶向去整合素金属蛋f t酶9(A D A M 9)抑制乳腺癌细胞的侵袭和迁移。方法采用RT-PCR测定检测乳腺癌癌组织、癌旁组织和乳腺癌细胞中miR-140-5P表达。将B T549细胞分为mimic组、miR-140-5p组、pLV-ADAM9组和pLV-ADAM9+miR-140-5p组,荧光素实验验证miR-140-5p和ADAM9靶向关系。检测各组细胞侵袭、迁移、细胞层粘连蛋白1(h m iin in l)、血管内皮生长因子(V E G F)、E钙黏附素(Ecadherin)、基质金属蛋白酶(MMP)-2蛋白表达。结果与癌旁组织相比,癌组织中miR-140-5p表达下调(P<0.01),乳腺癌BT549细胞中m iR-140-5p表达量最低;m iR-140-5p可以靶向负调控ADAM9的表达,抑制细胞侵袭、迁移(P<0.05),上调E cadherin蛋白表达(P<0.05),下调L am ininl、V EG F和M M P-2蛋白表达(尸<0.05)。结论m iK-140-5p可以靶向负调控ADAM 9的表达,抑制乳腺癌细胞的侵袭和迁移。
Objective To investigate how miR-140-5p inhibits the invasion and migration of breast cancer cells by targeting A Disintegrin and Metalloprotease 9(ADAM9).Methods RTPCR was performed to detect the expression of miR-140-5p in breast cancer tissues,para-carcinoma tissues,and and breast cancer cells.BT549 cells culturedi/i vitroy/ere divided into mimic group,miR-140-5p group,pLV-ADAM9 group and pLV-ADAM9+miR-140-5p group.The fluorescein assay was conducted to further verify the targeting relationship between miR-140-5p and ADAM9.Cell invasion,migration and the expression of laminin 1(Laminin 1),vascular endothelium growth factor(V E G F),E-cadherin(Ecadherin)and matrix metalloproteinase(MMP)-2 protein were detected.Results Compared with para-carcinoma tissues,expression of miR-140-5p was significantly down-regulated in cancer tissues(P<0.01).The expression of miR-140-5p was lowest in BT549 cells.The miR-140-5p could specifically down-regulate ADAM9 expression to inhibit invasion and migration of BT549 cells(P<0.05),up-regulate Ecadherin protein expression(P<0.05),down-regulate the expression of Lamininl,VEGF and MMP-2 protein(P<0.05).Conclusion The miR-140-5p can specifically down-regulate ADAM9 expression to inhibit invasion and migration of breast cancer cells.
