摘要
目的探讨Twist过表达对SKOV3细胞血管生成拟态和上皮间质转化的作用。方法选择上皮性卵巢癌和正常卵巢组织各20例,CD31-PAS双染检测上皮性卵巢癌组织中血管生成拟态的情况。SKOV3细胞分别转染Twist过表达载体和抑制载体后,应用流式细胞术分别检测SKOV3细胞表面血管内皮标志物CD31、CD34的表达;应用三维细胞培养技术检测SKOV3细胞的上皮间质转化;real-time PCR和Western blot检测E-cadherin的表达。结果上皮性卵巢癌组织中血管生成拟态显著高于正常卵巢组织(5.25±0.56 vs 1.10±0.45,P<0.05)。Twist过表达组CD31和/或CD34阳性的SKOV3细胞数目明显多于空白组、空载组以及Twist-shRNA782组(P<0.05);Twist过表达组SKOV3细胞的上皮间质转化远高于空白组、空载组和抑制表达组(6.83±0.49 vs 0.92±0.31,1.41±0.38,1.25±0.39,P<0.05);Twist过表达组E-cadherin的表达降低(P<0.05)。结论Twist能够促进SKOV3细胞的血管生成拟态和上皮间质转化。
Objective To investigate the effects of Twist overexpression on the vasculogenic mimicry and epithelial-mesenchymal transition of SKOV3 cells.Methods The vasculogenic mimicry was tested in 20 epithelial ovarian cancer tissues and 20 normal ovary tissues by CD31-PAS staining.SKOV3 cells were divided into four groups:Twist overexpression group,Twist inhibition group,negative control group and empty vector group.The expression of CD31 and CD34 in SKOV3 cells was tested by flow cytometry.The epithelial-mesenchymal transition in SKOV3 cells was detected by 3D culture.The expression of E-cadherin was detected by real-time PCR and Western blot.Results The vasculogenic mimicry of epithelial ovarian cancer tissues was higher than that of normal ovary tissues(5.25±0.56 vs 1.10±0.45,P<0.05).The positive rate of CD31 and/or CD34 was higher in Twist overexpression group than that in control group,empty vector group and Twist inhibition group(P<0.05).The epithelial-mesenchymal transition in SKOV3 cells was higher in Twist overexpression group than that in control group,empty vector group and Twist inhibition group(6.83±0.49 vs 0.92±0.31,1.41±0.38,1.25±0.39,P<0.05).Twist overexpression decreased the expression of E-cadherin(P<0.05).Conclusion Twist can promote the vasculogenic mimicry and epithelial-mesenchymal transition in SKOV3 cells.
