摘要
目的:探讨免疫组化室内质控中应用组织悬液作为HER-2(人表皮生长因子受体-2)阳性对照的效果,为建立更好的HER-2免疫组化室内质控提供参考。方法:择取2018年1月-2019年12月笔者所在医院的37例HER-2(3+)的胃癌根治标本,包埋成石蜡块连续切片,行免疫组化染色,另取切片搅碎后制成组织悬液,分别进行免疫组化染色和常规HE染色,于试验后4、12周时,复行免疫组化染色,比较观察染色定位及强度情况。结果:组织悬液标本HE染色镜下观察均可见异形癌细胞,多数平铺,轮廓清晰。免疫组化染色的胃癌组织悬液标本,HER-2表达均见于细胞膜,染色强度均为强阳性(3+),同石蜡切片定性定位与染色均相符。试验后4、12周复行免疫组化,HER-2染色定位与强度同前无明显改变。结论:组织悬液癌细胞HER-2阳性定位明确,染色强度准确,质量稳定易判读,能够保障染色过程可靠,可以作为免疫组化室内质控,为HER-2染色提供阳性参考。
Objective:To investigate the effect of using tissue suspension as a positive control of HER-2(human epidermal growth factor receptor-2)in the quality control of indoor immunohistochemistry,and provide a reference for establishing a better quality control of HER-2 immunohistochemistry indoor.Method:From January 2018 to December 2019,37 radical specimens of HER-2(3+)gastric cancer in our hospital were selected and embedded into paraffin sections for immunohistochemical staining,and then the sections were broken to make tissue suspension.Immunohistochemical staining and routine HE staining were carried out respectively.At 4 and 12 weeks after the test,immunohistochemical staining was carried out again to observe the location and intensity of staining.Result:The heteromorphic cancer cells were observed under HE staining microscope,most of them were flat and clear.The expression of HER-2 was found in the cell membrane of gastric cancer tissue with immunohistochemical staining,and the staining intensity was strong positive(3+),which was consistent with the qualitative localization and staining of paraffin section.After 4 and 12 weeks of the experiment,immunohistochemistry was carried out again,and the location and intensity of HER-2 staining were the same as before.Conclusion:HER-2 positive localization of cancer cells in tissue suspension is clear,the staining intensity is accurate,the quality is stable and easy to read,which can guarantee the reliable staining process,and can be used as the indoor quality control of immunohistochemistry,providing positive reference for HER-2 staining.
