摘要
目的:观察梓醇对AAPH损伤后少突胶质细胞前体细胞(OPC)突起的保护作用。方法:神经干细胞(NSC)诱导产生OPC,使用不同浓度时间AAPH损伤OPC,选取合适损伤时间浓度的AAPH,建立OPC损伤模型。利用不同浓度梓醇对损伤OPC进行干预,CCK-8法测定细胞存活率,Olig1鉴定及观察OPC突起损伤程度,计算OPC细胞突起率。结果:和空白对照(NC)组细胞比较,模型(MO)组细胞CCK-8吸光度显著降低,细胞突起率明显减少(P<0.01),Olig1积分光密度显著降低。与MO组比较,不同浓度梓醇均能显著增加OPC细胞的存活率(P<0.05,P<0.01),保护OPC细胞突起,增加Olig1蛋白的表达,其中,以10μmol/L浓度梓醇效果最佳。结论:梓醇对AAPH损伤后的OPC具有显著的保护作用。
Objective:To observe the protective effect of catalpol on oligodendrocyte premature cell(OPC)protrusion after AAPH injury.Methods:Neural stem cell(NSC)induced OPC,OPC was used to damage AAPH at different concentration times,and AAPH with appropriate injury time concentration was selected to establish OPC injury model.Different concentrations of sterol were used to intervene the injured OPC.CCK-8 method was used to determine the cell survival rate.Olig1 was used to identify and observe the OPC protrusion damage degree.The OPC cell protrusion rate and the Olig1 integrated optical density were calculated.Results:Compared with the blank control(NC)group cells,the CCK-8 absorbance of the model group(MO)cells,the cell protrusion rate,and the Olig1 integrated optical density significantly decreased(P<0.01).Compared with MO group,different concentrations of sterol could significantly increase the survival rate of OPC cells(P<0.05,P<0.01),protect OPC cell processes and increase the expression of Olig1 protein.Among them,the best concentration of catalpol was 10μmol/L.Conclusion:Sterol has significant protective effect on OPC after AAPH injury.
