摘要
目的在低钾任氏液灌注状态下观察茶色素对离体蟾蜍心脏的心率、心肌收缩力和心电图的影响。方法80只蟾蜍随机分为对照组和实验组。对照组包括任氏液组和低钾任氏液组(K+浓度分别为0.07、0.035、0.0175 mg/mL),实验组为低钾任氏液(K+浓度分别为0.07、0.035、0.0175 mg/mL)+茶色素(200、400μg/mL)组。制备离体蛙心灌流标本采用斯氏法,运用BL-420S生物机能实验系统同时记录离体蟾蜍心肌收缩力和心电图曲线,进行统计学的方法分析。结果蟾蜍离体心脏在低钾任氏液灌注时可出现心率显著下降(P<0.05),心肌收缩力显著减弱(P<0.05),心电图异常如出现P-R间期延长、量子共振检测(quantum resonance spectrometer,QRS)综合波增宽、出现明显U波等。茶色素可使在低钾任氏液灌注后的离体蟾蜍心脏心肌收缩力和心率的减弱显著增强(P<0.05),异常的心电图恢复正常。结论茶色素可以影响低钾状态下的离体蟾蜍心脏低下功能,并可恢复至正常。
Objective To investigate the effects of tea pigment on heart rate,myocardial contractility and electrocardiogram of isolated toad heart under the condition of low potassium Ren’s solution perfusion.Methods Eighty toads were randomly divided into control group and experimental group.The control group included Ren’s solution group and low potassium Ren’s solution group(K+concentration was 0.07,0.035,0.0175 mg/mL respectively),while the experimental group was low potassium Ren’s solution(K+concentration was 0.07,0.035,0.0175 mg/mL respectively)+tea pigment(200,400μg/mL).The preparation of isolated frog heart perfusion standard was carried out by Stephen’s method,and the contractility and electrocardiogram curves of isolated toad heart were recorded simultaneously by BL-420 S biological function experimental system,and statistical analysis was carried out.Results The heart rate of toad isolated heart decreased significantly(P<0.05),myocardial contractility decreased significantly(P<0.05),ECG abnormalities such as prolonged P-R interval,widened QRS complex wave and obvious U wave appeared when perfused with low potassium Ren’s solution.Tea pigment could significantly enhance the weakening of myocardial contractility and heart rate of isolated toad heart perfused with low potassium Ren’s solution(P<0.05),and abnormal electrocardiogram returned to normal.Conclusion Tea pigments can restore heart function to normal under low potassium condition.
